Objective This study was purposed to investigate the growth inhibition and telomerase effect of3’-azido-2’,3’-dideoxythymidine (AZT) on the human acute myeloid leukemia cell line KG-la.Methods The effect of proliferation was detected by MTT assay after the KG-1a cell were stimulated for24h,48h,72h with different concentrations of AZT;Cell cycle and cell apoptosis was detected by Flow Cytometry;Telomerase activity was detected with TRAP-PCR-ELISA assay; Telomerase hTERT-mRNA expression detect by RT-PCR.Results The proliferation of KG-1a cells was inhibited in a time and dose dependent manner; The KG-la cells in S phase decreased and in G2/M phase increased compared with control group (p<0.05). Telomerase activity and hTERT-mRNA expression in the experimental group decreased than those in control group (p<0.05), the lower degree was positilvely correlated with concentration of AZT.Conclusion AZT inhibits KG-1a cells proliferation and induces apoptosis, which maybe related with its decreasing the telomerase activity and hTERT-mRNA expression.
