Effect Of AZT On Proliferation, Apoptosis And Telomerase Activity Of KG-1a Cells | Posted on:2013-12-21 | Degree:Master | Type:Thesis | Country:China | Candidate:R R Jin | Full Text:PDF | GTID:2234330371987389 | Subject:Internal Medicine | Abstract/Summary: | PDF Full Text Request | Objective This study was purposed to investigate the growth inhibition and telomerase effect of3’-azido-2’,3’-dideoxythymidine (AZT) on the human acute myeloid leukemia cell line KG-la.Methods The effect of proliferation was detected by MTT assay after the KG-1a cell were stimulated for24h,48h,72h with different concentrations of AZT;Cell cycle and cell apoptosis was detected by Flow Cytometry;Telomerase activity was detected with TRAP-PCR-ELISA assay; Telomerase hTERT-mRNA expression detect by RT-PCR.Results The proliferation of KG-1a cells was inhibited in a time and dose dependent manner; The KG-la cells in S phase decreased and in G2/M phase increased compared with control group (p<0.05).
