| 1 The objectivesThe Radix Fici Hirtae is the dry root of Ficus hirta Vahl.from mulberry branch plant,it has a good effect for the spleen dispel the wet,benefits the lung to relieve a cough,stretch the muscle and bone.Commonly it is used in the sickness about spleen weakness and edema,losses of appetite and incapable,hepatitis,rheumatism pain,the cough for pulmonary tuberculosis and so on.Also it is used in brewing alcohol,making soup and tea,It is not only the drugs and also the food.It is one of development potential resources.This topic objectives of the research can be illustrated as follows, the first one is to discuss the function of The Radix Fici Hirtae to make up the spleen weakness,provides the rationale for the clinical utilization.The second one is to observe the acute toxicity and the Maximal tolerance dose(MTD) of The Radix Fici Hirtae in mice respectively for establish nontoxic dose to providing reference in clinical medication safety.The third one is to establish fingerprint atlas by HPLC and Quality control atlas by TLC about The Radix Fici Hirtae which is from Heyuan area.2 The methodsThis research is mainly divided two parts.One part is the literature search including the domestic and foreign literature as the foundation of the research.Another part is the experimental study,including:Main pharmacodynamics research,acute toxicity and the Maximal tolerance dose experimental study,Quality control atlas research.(1) Through acute toxicity test and Maximal tolerance dose experimental study,to observe the death,the activity and the diet situation of mice, and through anatomizing the internal organs to observe exceptionally of the heart,liver,spleen,lung,kidney,pancreatic gland,small,intestine and so on,and the pathology inspection determined whether the Radix Fici Hirtae is noxiously or not.And give the max for dose.(2) Uses Rheum palmatum L.to manufacture the spleen weakness card model mice,Observes the effect about the Radix Fici Hirtae to the spleen weakness model mice.From the follow target:the Mice's general symptom change,the grows of body weight,the spleen index and the thymus index,activeness of serum amylase,the stomach' s excretion speed,the speed of the intestines advances,the time exist in the anoxic environment,capability of resisting the cold and fatigue.(3) Through the HPLC atlas and TLC method to control the quality of the Radix Fici Hirtae which is planted in heyuan area.3 Results3.1 Literature search resultThe literature research indicated that,the most research of the Radix Fici Hirtae is about its chemical composition,few about its pharmacological action research.In particular there is on study about the pharmacological action of regarding makes up the spleen.The related toxicity is always the experiential know,and does not have the tentative data support.Some literature reported that the medicinal materials' quality from heyuan area is better than anther area,and content of the effective component psoralea corylifolia element is higher.Therefore it has the necessity to carry on the research for medicine to control the atlas.3.2 Experimental study result(1) The pharmacodynamics experimental study indicated that,the Radix Fici Hirtae can enhancement the content of the spleen weakness model mice' s serum amylase(P<0.05),can determinately restrain the speed of the model mice' s stomach evacuation and the small intestine(P<0.05-0.01),can lengthen the survival time of the model mice' s under the environment of atmospheric pressure and anoxic(P<0.01-<0.05),can increase the model mice' s thymus weight(P<0.05),can lengthen swimming time of the model mice under low temperature condition(P<0.05),also can increase the model mice' s body weight(P<0.01).Above all affords the theory support about the Radix Fici Hirtae' s restorative function of spleen weakness. (2) The result of the acute toxicity test shows that,after 3days there is no death of the mice which are gave the medicine,the activity and the dietetic situation are normal.The activity of the mice is slow-down slightly in the Maximal tolerance dose experimental study,and restores normally after 30min.There is no death of the mice which are gave the medicine after 14days.And the mice' s eyes noses mouths have non-mucous secretion.The statistics analysis result of the mice body weight,potable water quantity and the feed quantity not remarkable difference(P>0.05). The most greatly use dosage is 370 times of the adult everyday dose.The organs'(includes heart,liver,spleen,stomach,lung, kidney,pancreatic gland,large intestine,small intestine and so on.) outward appearance,the luster and the size do not have exceptionally.The medicine group mice' s internal organs of pathology inspection has not seen the change of medicine pathology.(3) The result of HPLC shows that there' re 13 peaks of the 9 samples in the HPLC fingerprint atlas.The similarity of them is higher than 0.9. It is explained that the chemical composition of the medicine which are from heyuan area is similar.The quality of the Radix Fici Hirtae is all stable never the wild or the GAP base planter.(4) The results of TLC shows that the characteristic spots on the chromatogram of the preparation were corresponding to the ones of contrast product.It explains that the psoralea corylifolia element be separated from the original medicine.The characteristic spots which demonstrated in the chromatograph are few,says that the impurity from extraction by methyl alcohol are few compare to aether.And separation of the characteristic spots is better.4 Research conclusionThrough the pharmacodynamics experiment indicated the Radix Fici Hirtae has the good function to enhance spleen weakness mice' s each target,provides the rationale for the function of makes up spleen weakness symptom.Moreover it has the Merit of high secure,the test pharmacology security result prompts that it is nearly no toxicity in the normal oral administration dosage situation.There is extremely slight effect to(?)organism' s internal organs even no influence. The result of the fingerprint atlas experimental study shows that this HPLC method is simple,rapid,accurate and reproducible,and can be used for the quality control of the Radix Fici Hirtae which is from heyuan eara.This TLC method has good dissociation specialization,reproducibility, and sensitivity.It can be used for the quality control of the Radix Fici Hirtae which is from heyuan eara.The part of Pharmacodynamics research is not yet comprehensive,for example if whether the psoralea corylifolia element is the effective component cannot be determined.
