The regenerated Antheraea yamamai/Bombyx mori(A.y/B.m) silk fibroin blend film and porous material were prepared by blending regenerated Antheraea yamamai and Bombyx mori silk fibroin solution.This paper took a research on the relationship between preparation conditions and physic-chemical properties.The cytocompatibility and histocompatibility were also studied.The A.y/B.m blend films were prepared by casting A.y/B.m mixture solution on a polyethylene plate and air-dried.The condensed structure of blend films was characterized by XRD,FT-IR and Raman spectra.The physical properties such as broken strength,initial tensile modulus and broken elongation in wet state were analyzed.The L929 fibroblast cells were cultured on the blend films to examine the cytocompatibility.The results indicated that there were rather bad compatibility between A.yamamai silk fibroin and B. mori silk fibroin.The blend films after ethanol treatment showed phase separation microstructure and no obvious effects took place on the molecule segment of these two kinds of silk fibroin.There were mainlyα-structure and random coil in A.y/B.m blend films before ethanol treatment.After treated by 80%ethanol,the condensed structure changed toβ-sheet structure.Comparing with the A.yamamai silk fibroin film,the mechanical properties and flexibility of blend film were improved.A.yamamai and B. mori silk fibroin might synergistically facilitate the cell proliferation;therefore,the blend film had good cytocompatibility.A.y/B.m blend porous materials were prepared by freeze-drying.The condensed structure of blend porous materials and pore structure were characterized.It found that two different kinds of silk fibroin in porous material had inferior compatibility and the interaction force was rather poor.The condensed structure of A.y/B.m blend porous materials before ethanol treatment coexists withα-structure and random coil.After treated by ethanol,the structure is mainlyβ-sheet structure.The pore diameter and porosity decreased with the increasing of proportion of B.mori silk fibroin and the falling of freezing temperature,while the pore density had opposite tendency.By adjusting the technical condition,the A.y/B.m blend porous materials could be prepared with the average pore diameter from 92μm to 320μm,the porosity from 78%to 92%.The A. yamamai silk fibroin porous materials were implanted into the skin defect in sprague-dawley(SD) rat to evaluate the histocompatibility.Compared with PVA scaffold, the expression ofPCNA,VEGF,α-SMA,Col-Ⅰ,Col-Ⅲ,TGF-β1,bFGF-2 in silk fibroin are rather high,however,there are no significant differences of expression of CD90 between of these kinds of materials.The results suggested that silk fibroin porous materials could guide the formation of functional microvessels and induce the regeneration of dermal tissue.The silk fibroin scaffold would be biodegradation gradually during the regenerative process.
