| Objective:Establish the pattern of Balb/C mouse skin's photo-damage to observe the effect of gypenosides(GP) on photo-damage and identify the correlation between gypenosides and anti-photo-damageMethods:we set up a total of 10 groups Blab/C mouse skin photo-damage's model by 7 times UVB irradiation.Before Brad-Ford be used to detect the protein.The nuclear NF-KB Inhibitor(IKB) protein as well as inhibition KB of protein kinase(IKK) expression were detected by western blotting.Groups as follow:Control groups,UVB irradiation group,UVB + Matrix groups,Matrix groups + UVB,UVB + vitamin E cream groups,vitamin E cream groups + UVB,UVB +1.5%Gypenosides groups, 1.5%groups Gypenosides +UVB,UVB +6%Gypenosides groups,6%Gypenosides + UVB groups.Results:1) IκB,IKK protein did not expressed in the control groups.2)For UVB irradiation groups the expression of IκB was significantly decresed compared to other groups,and IKK protein expression higher than other groups.3) Application 1.5%concemtration of gypenosides groups and 6%concemtration of gypenosides groups the protein of IκB expression was significantly higher than UVB irradiation groups,whereas IKK protein expression was significantly lower than UVB irradiation groups,simila with the positive control group of vitamin E.4)Compared with UVB irradiation groups IκB protein expression had no significant increase in the matrix groups,but the matrix of the IKK protein expression had decreased.5) Pretreate vitamin E groups and 1.5%cream Gypenosides Groups IκB protein expression higher than UVB exposure.6) Pretreate VitE groups and 1.5%cream Gypenosides Groups IKK protein expression lower than UVB exposure Conclusions:1) UVB can significantly inhibit IKB protein and increas IKK protein expression of the Balb / C mouse skin tissue,thus activate of NF-KB(NF-kappa B) by UV radiation.2) 1.5%Gypenosides cream groups and 6%Gypenosides cream groups can enhance IkB protein expression of the Balb / C mouse skin tissue by UVB exposure,re-establish of UVB-induced degradation of IkB protein expression;IKK protein expression activity is inhibited,thereby inhibiting the activity of NF-KB, those role are similar to the positive control group of vitamin E cream,may be one of the ways of anti-inflammatory and anti-photo-damage role.3) Matrix cream groups can not increase IkB protein expression of the Balb / C mouse skin tissue by UVB exposure。4) VitE cream groups,1.5%cream Gypenosides Groups piror to UVB exposures resulted in a further enhancement in UVB-induced degradation of IkB protein expression;IKK protein expression activity is inhibited... |
