Objective Study of psoriasis patients with bone marrow-derived mesenchymal stem cell(BMSCs) isolated and cultured in vitro,identification and cell growth conditions, culture supernatants were measured the level of cytokine secretion and cytokine associated with disease activity of analysis,so as to explore the pathogenesis of psoriasis.Metheds MSCs were isolated from bone marrow of both 24 patients and 20 healthy controls by density gradient centrifugation and adhensive cultrure in vitro.To identify cell surface markers by flow cytometry.Through the inverted phase contrast microscope to observe morphological changes of BMSCs,the situation of the growth and adhesion, We detected the expressions of IL-6,VEGF,TGF-βof cell culture supernatants by ELISA.Psoriasis area and severity of the rash using PASI score method,the expression of cytokines and its relationship with disease activity using pearson correlation analysis.Results Successfully isolated and cultured to obtain primary cultured BMSCs and to the 4th generation,flow cytometry showed that BMSCs expressed surface antigens CD29,CD44 and CD34,CD45,CD106,HLA-DR expression is negative.BMSCs in patients with psoriasis compared with the control group affixed to the wall later and a state of slow growth..IL-6 concentration of BMSCs culture supernatant was significantly higher than normal(P<0.01),VEGF significantly lower than normal concentrations(P<0.01),BMSC secretion of TGF-βlevels and the control group no significant difference (P>0.01).IL-6,VEGF and TGF-βlevels of Patients and PASI score had no correlation (P>0.05).Conclusion Cytokine IL-6,VEGF secretion of BMSCs in patients with psoriasis are abnormal,indicating that bone marrow hematopoietic microenvironment of psoriasis are abnormal,and thus the development of psoriasis hematopoietic stem cells may have an impact.That psoriasis patients with bone marrow hematopoietic microenvironment and hematopoietic stem cell abnormalities may be involved in the pathogenesis of psoriasis.
