Purpose:To study the relation between the cyclin D1(CCND1) A870G gene polymorphism and age,sex,smoking,family history,histology,tumor stage and the outcome of chemotherapy for the patients with non-small cell lung cancer(NSCLC) in YunNan.Methods:Cyclin D1(CCND1) A870G gene polymorphism was determined by polymerase chain reaction(PCR) and restriction fragment length polymorphism analysis(RFLP) of DNA which extracted from blood. The study included 300 non-small cell lung cancer(NSCLC) patients and 500 healthy control subjects in Yun Nan province.Use theχ~2 inspection analyze the relationship between the distributional difference of each genotypic frequency and the clinical parameter of sex age stage of pathology and so on,calculation odds ratio (OR) of relative risk(RR) and 95%confidence interval(CI).Results:The A,G allele frequencies between non-small cell lung cancer(NSCLC) patients and healthy controls was 74.7%,81%and 75.8%,80.8%,There were no significant differences(P=0.907>0.05).The AA,AG and GG genotype frequencies were 19%,25.3%,55.7%respectively in non-small cell lung cancer patients and 19.2%,24.2%,56.6%in healthy controls.The AA,GG and AG genotype frequencies in cyclinD1(CCND1) A870G of patients and controls both conformed to the Hardy equilibrium.The GG genotype frequencies in the patients who have the history of heavy smoking was significantly higher than that of GG and AA(≥40py,P =0.034<0.05).The adjusted odd Ratio was 2.124,95%CI=1.067~4.228.The AA genotype frequencies of the adenocarcinoma patients was significantly higher than that of GG and AA(P=0.019<0.05).The adjusted odd Ratio was2.022,95% CI=1.121~3.649.In the grope of CR/PR in GP first-line chemotherapy,The GG genotype was significantly higher than that of AG and AA(P = 0.001<0.05).The adjusted odd Ratio was 8.333,95%CI=2.255~30.796.Conclusion:Evidence was obtained that the GG genotype of the CCND1 A870G gene polymorphism modulates smoking-induced lung cancer risk.and AA genotype of CCND1 A870G gene polymorphism modulates adenocarcinoma risk.The patients with GG genotype of CCND1 A870G gene polymorphism can be sensitivity for GP and GG genotype can be as a predictor for GP sensitivity in NSCLC patients.
