ObjectiveTo investigate the therapeutical effect of Shexiang Baoxin Pills and mobilization of autologous bone marrow stem cells (ABMSCs) for myocardial infarction (MI), and de- montrate the mechanism of Shexiang Baoxin Pills and granulocyte colony-stimulating factor (G-CSF) for influencing heart function after MI.MethodsThe model of MI was created by ligating left antierior descending (LAD) coronary artery in rabbits which were divided into four groups that were control group (groupâ… ), group of ABMSCs mobilized by G-CSF (groupâ…¡), Shexiang Baoxin Pills group (groupâ…¢), and the group of Shexiang Baoxin Pills based on G-CSF ABMSCs mobilized by G-CSF (groupâ…£). General state of health was monitored after operation. Left ven- tricular ejection fraction (LVEF), left ventricular end-diastolic dimension (LVEDd) and left ven- tricular wall motion (LVWM) were detected by ultrasound-cardiogram (UCG) before operation, one week after operation and six weeks after operation. The hearts were harvested for pathologic detection after six weeks. Capillary density (CD) at centre and marginal zone of infarct myocardium was monitored, and the infarct size of myo- cardium was calculated.Results1. Compared to groupâ… , the general state of the animals took a favorable turn in groupâ…¡,â…¢andâ…£after operation. 2. One week after operation, LVEF was 50.98±6.78% in groupâ… , 53.72±4.91% in groupâ…¡, 54.46±4.58% in groupâ…¢and 54.83±5.92% in groupâ…£. LVEDd was 17.77±1.59mm in groupâ… , 17.02±1.64mm in groupâ…¡, 17.15±1.89mm in groupâ…¢and 17.19±1.71mm in groupâ…£. Six weeks after operation, LVEF was 50.89±8.32% in groupâ… , 56.43±5.77% in groupâ…¡, 57.25±6.74% in groupâ…¢and 60.38±4.43% in groupâ…£. Variation of LVEF was -0.09±2.78% in groupâ… , 2.71±3.64% in groupâ…¡, 2.79±3.74% in groupâ…¢and 5.55±3.88% in groupâ…£. LVEDd was 17.71±1.64mm in groupâ… , 15.97±1.52mm in groupâ…¡, 16.17±1.26mm in groupâ…¢and 15.76±1.30mm in groupâ…£. Variation of LVEDd was 0.06±0.69mm in groupâ… , 1.05±1.26mm in groupâ…¡, 0.98±1.40mm in groupâ…¢and 1.43±1.55mm in groupâ…£. Compared to one week after operation, six weeks after operation, LVEF and LVEDd had no marked change (P>0.05) in groupâ… , but in other three groups, LVEF improved significantly (P<0.05), LVEDd decreesed (P<0.05) and LVWM enhanced sign- ificantly. Six weeks after operation: (1) Group comparison in LVEF: LVEF in Groupâ…£had obvious difference with it in groupâ… andâ…¡(P<0.05), but had no difference with it in groupâ…¢(P>0.05). LVEF in Groupâ…¡andâ…¢had no difference, but both increased more than groupâ… (P<0.05). (2) Group comparison in variation of LVEF: Variation of LVEF in groupâ…£had significant deviation with it in groupâ… andâ…¡(P<0.05), but had no difference with it in groupâ…¢(P>0.05). Variation of LVEF in groupâ…¡andâ…¢had no difference, but both had statistical significance with it in groupâ… (P<0.05). (3) Group comparison in LVEDd: LVEDd in groupâ…¡,â…¢andâ…£had no statistical significance with each other (P>0.05), but all had obvious difference with it in groupâ… (P<0.05). (4) Group comparison in variation of LVEDd: Variation of LVEDd in Groupâ…¡,â…¢andâ…£had no significant deviation with each other (P>0.05), but all had obvious difference with groupâ… (P<0.05).3. The number of blood capillary at marginal zone of infarct myocardium was 13.69±2.93 in groupâ… , 22.87±2.70 in groupâ…¡, 20.47±2.90 in groupâ…¢and 25.00 ±2.54 in groupâ…£. The number of blood capillary at centre of infarct myocardium was 2.54±1.27 in groupâ… , 6.20±1.26 in groupâ…¡, 4.33±1.54 in groupâ…¢, 8.33±1.95 in groupâ…£. Group comparison in CD at marginal zone and centre of infarct myocardium: The number of blood capillary in groupâ…£was more than in other three groups. It was the slightest in groupâ… . There was more blood capillaries in groupâ…¡than in groupâ…£(all P<0.05).4. Infarct size of myocardium was 37.15±2.94% in groupâ… , 27.47±2.13% in groupâ…¡, 28.33±2.35% in groupâ…¢, 26.73±1.83% in groupâ…£. Group comparison in infarct size of myocardium: It was smaller in groupâ…£than in groupâ… andâ…¢(P<0.05), but had no statistical significance with it in groupâ…¡(P>0.05). It had no difference in Groupâ…¡andâ…¢(P>0.05), but both smaller than it in groupâ… (P<0.05).ConclusionsShexiang Baoxin Pills, ABMSCs mobilization by G-CSF and Shexiang Baoxin Pills based on ABMSCs mobilization by G-CSF could improve general state of rabbits. All of them could increase CD, decrease infarct size of myocardium and improve heart function of rabbits with MI. There was an improvement in LVEF with Shexiang Baoxin Pills based on mobilization of ABMSCs by increasing CD and decreasing infarct size further, but no obvious changes in LVEDd.
