Objective:To study the relationship between coagulation indexes PT(Plasma Prothrombin Time),APTT(Activated partial thromboplastin time),PLT(Platelet),FIB(Fibrinogen), DD(D-dimer),vWF(von Willebrand factor) and malignant tumor metastasis.Methods:1.120 malignant tumor patients were observed,including 16 colon cancer,19 ovarian cancer,22 pancreatic cancer,25 gastric cancer,12 prostatic cancer,17 lung cancer,9 urinary bladder cancer.Sysmex CA7000 automated multiparameter hemostasis analyzer was used to detect the plasma levels of PT,APTT and FIB with clotting method,to detect DD level with immunoturbidimetry and vWF with immunologic assays.The plasma PLT was counted by Sysmex XE2100 haematology automated analyzer.The results were compared with those of health controls.2.In vitro,human megacell lung cancer NCI-H460 was cultured.The adhesion of NCI-H460 cells to vWF was observed by MTT method;The effect of Anti-vWF on tumor cells NCI-H460 adhesion to vWF was also observed by MTT.Results:1.There are significant difference of the plasma levels of PT,APTT,PLT,FIB,DD, vWF between malignant tumor group and health control group(P<0.05);There are significant difference of the plasma levels of PT,APTT,PLT,FIB,DD between malignant tumor without metastasis group and health control group(P<0.05),but there are no significant difference on the plasma level of vWF between these two group;There are significant difference of the plasma levels of FIB,DD,vWF between malignant tumor with metastasis group and malignant turnor without metastasis group(P<0.05),but there are no significant difference on the plasma level of PT,APTT,PLT between malignant tumor groups with/without metastasis. 2.Human megacell lung cancer NCI-H460 can adhere to vWF at different concentration.The OD of vWF group at different concentrations(1μg/ml,2μg/ml, 3μg/ml,4μg/ml) were significant higher than control group(P<0.05),and the higher concentrations of vWF were,the higher OD value were.When the concentrations of vWF reach 2μg/ml,the OD value didn't increase with the raise of vWF level.There are no significant difference of OD value among vWF groups at different concentrations(2μg/ml,3μg/ml,4μg/ml).Anti-vWF(1μg/ml,1.5μg/ml,2μg/ml,2.5μ/ml ) can significantly inhibite NCI-H460 cells adhesion to vWF in vitro(P<0.05),and the higher concentrations of Anti-vWF were,the higher inhibition were.The highest inhibition rates are 65.05%.When the concentrations of Anti-vWF reach 2μg/ml,the inhibition didn't increase with the raise of Anti-vWF level.There are no significant difference of OD value between 2μg/ml Anti-vWF group and 2.5μg/ml Anti-vWF group.Conclusion:The higher of PT,APTT,PLT,FIB,DD,vWF on malignant tumor patients indicate that blood hypercoagulabale state exists.The detection of these indexes may play a specific role in early discovery of hypercoagulabale state and hypercoagulabale state tendency on malignant tumor patients,and provide standards for the laboratory examination.According to the detection of these indexes,maybe we can indirectly prognosticate neoplastic occurrence,infiltration and transfusion.
