【Objective】To establish rat model of partial hepatectomy(PH) with local outflow obstruction and investigate effect of local outflow obstruction on regeneration alter partial hepatectomy in rats.【Methods】Part1:20 healthy aldut male Wistar rats were divided into two groups:Ten rats as experimental group were performed with 50%PH;afterwards,their right of median vein(RMHV) were ligated.Another ten rats as control group received 50%PH only.Long-term survival and stability of the two groups were compared.Part2:48 healthy aldut male Wistar rats were divided into two groups: Experimental group were performed with 50%PH;aflerwards,their RMHV was ligated.The control group received 50%PH only.The width of portal vein was measured during the operation.6 rats of each group were killed at 1,2,3,7 day after operation.Draw blood from inferior vena cava to detect ALT;Count liver to body rate, perform regular pathology for liver and count mitosis index of hepatic cell,immunohistory chemistry for proliferating cell nuclear antigen(PCNA),flow cytometry for cell cycle to count proportion of S phase.【Results】Part1:All the ten rats suffered local outflow obstruction after their right veins of median lobe were ligated;about 1/3 of median lobe occurred congestion on the right;both of the two groups survived without any death within one month after operation.Part2:The rate of change in portal vein of the experimental group were higher than the control group.ALT of experimental group were higher than the control group in the early period;the outflow obstruction area of the experimental group occoured necrosis and finally becomed atrophy;activation was seen in the portal area on the first day after operation,the.liver regeneration peak of the other normal area also delayed compared with the control group.【Conclusions】1.The rat model of partial hepatectomy with local outflow obstruction is stable whose success rate is 100%could be used for related research.2.Hepatic cell in outflow obstruction area infracted,were absorbed and finally atrophy;and no regeneration occur.3.Local outflow obstruction delayed liver regeneration aider partial hepatectomy in rats in some degrees and caused hepatic fuction damaged in the early period;its mechanism may be related to portal hypertension and activation of Kuffer cell in the obstruction area.
