Part One: establishment of liver regeneration model after partial hepatectomy in mice.Objective: to establish a liver regeneration(PH)model after 70% of liver resection in mice,which will lay a foundation for the study of stromal organization for liver regeneration.Methods: 12 male mice were divided into four groups,preparing the model of 2/3 hepatic resection of mice.They were divided into Sham group,PH 24 h group,PH 72 h group and PH 1w group,representing the liver regeneration the starting stage,stage of proliferation and termination stage.Meanwhile,serum and urine(for the standby application of molecular markers and metabolic groups)were collected.Ended up with the operation of 0 h,kill animals according to the request of grouping time points respectively in postoperative 24 h,7 h,7d.Weigh and record the weight of regenerative liver of each group.The liver tissue of each group was stained with Sirius scarlet to analyze the changes of ECM in each stage of liver regeneration.Results:1.12 mice all survived.Except the sham group,the other three groups showed different degree of liver regeneration.In this experiment the liver weight than of mice increased gradually over time.Until 24 h after surgery,the mouse liver weight can be restored to the original 40%.Until 72 h after surgery,the mouse liver weight can be restored to the original 70%.Until 7 days after surgery,the mouse liver weight can be restored to the original 90%.2.Sirius scar staining showed that in the PH24 h group,the disorder of the extracellular matrix and the decrease of the density were observed in the liver tissue.In the PH72 h group,the extracellular matrix increased more than before,and the arrangement was slightly more orderly than before.In the PH7 d group,the arrangement of the extracellular matrix was restored to normal.Conclusions: the liver regeneration model after 2/3 hepatic resection was successfully constructed by removing the left lobe and middle lobe of the mouse,and the liver size was basically in line with the process of liver regeneration.Part Two: extracellular mechanism of group study of liver regeneration after partial hepatectomy in mice.Purpose: to perform group study on the model of liver regeneration after partial hepatectomy in mice.The differential Matrix histone ECM m RNA gene was identified in each phase of liver regeneration after partial hepatectomy in mice.The expression and dynamic changes of Matrix histone ECM m RNA expression in liver regeneration were studied within the "whole" matrix group.This will lay the foundation for the multi-omics analysis of the matrix group for liver regeneration after the completion of the proteome study.Methods: by screening the m RNA expression profile,further determine the differential m RNA associated with the extracellular matrix of liver regeneration,and perform GO analysis on the differential genes associated with the extracellular matrix of liver regeneration in order to describe the functions of these genes.And by the Pathway analysis of differential genes,looking for if there is any relationship between the differential genes of different samples and the changes of any cellular pathways.Results: Screening differential m RNAs across groups within the matrix group,Compared with sham group,PH24 h group had 103 differentially expressed genes between the two groups,among which there were 58 up-regulated m RNAs,respectively Adam15;down-regulated m RNAs were 55;PH72h group compared with sham group,differentially expressed matrix group between the two groups There were a total of 103 protein m RNAs,including 7 collagens and 28 differential regulators.Compared with the PH24 h group,PH72 h group had a total of 19 differentially expressed matrix histone m RNAs,including 4 regulatory factor m RNAs,and differentially expressed glycoprotein m RNAs.Four of them had differentially expressed proteoglycans,one differentially expressed accessory protein,and eight differentially expressed matrix histone m RNAs were secreted factors.Compared with sham group,PH7 d group had 115 differentially expressed matrix histone m RNAs,among which 91 were Col4a1 up-regulated,down-regulated to 24 such as Adam1 a,and compared with PH72 h group,there were 36 ECM protein m RNAs differentially expressed in PH7 d group.There are 7 regulatory factor m RNAs.Conclusion: in the liver regeneration model,a large number of Matrix histone m RNA related to extracellular matrix are screened,which may play an important role in the regulation of liver regeneration stages.
