| Recurrent spontaneous abortion is most common seen complication during pregnancy,it jeopardizes reproductive health seriously.The etiology of RSA could be known in most cases,but there are quite a few still don't know why.So far we recognize the maternal immunity could response to the fetal antigen with Th2 induction.But if the response was deviated to Th1,the spontaneous happens.The notion of the Th2 epistatic immunity associated with the outcomes of pregnancy is recognized in the worldwide.If we can reverse the immune response from Th1 to Th2,therefore changing the harmful state.With modified bromocriptine-induced abortion model,the abortion rate is about 75~80%and the model was characterized by low serum PRL,and P level,and the immune response to Th1.according to the Chinese medicine theory that Bu-shen is closely retaliated to the reproductive health.RSA was correlated to shen-qin insufficient.Based on the theory,the effective of Bu-shen treatment is successful.lots of research indicated that shen theory based on Chinese medicine is correlated with immunity function.And lots of clinical research and animal experiment support the concept of Bu-shen,moreover Bu-shen treatment induced the immune response to Th2 immunity in rats experiment.Recently,SOCSs family(Suppressors of Cytokine Signaling) were just found,the function and structure of SOCS1,SOCS2,SOCS3 are clearer than other members of this family.SOCSs family is involved with Th cell induction and the transduction by many signals pathway,regulate the balance of cytokines,so that the importance of the SOCS is quite clear.And we assumed the SOCS might be necessary to the cytokines balance and the Th2 immunity in materal-fetal interface.With the recent progress of the founding of SOCSs family,we would like to investigate Bu-shen herbs treat the bromocriptine-induced abortion model at ducidua in maternal-fetal immunity affected SOCS expression.Objects:To study the effect of Chinese herbs Bu-shen of SOCS genes expression in associated with Th2 cytokines induced development in maternal-fetal interface. Material and methods:1.Animals:a total of 69 unpregnancy SD rats,aged 10~12 weeks,adult female rats weighed about 250~300g.Female and male rats were caged at 2:1.The day a sperm was found in vaginal smear was designated as day 1 of pregnancy,on day 6 to 8 pregnancy rats were injected with bromocriptine2.Female SD rats were divided into six groups randomly.Group A,B,C and D were injected with bromocriptine[0.3mg/(kg.d)]during the pregnant day 6~8,respectively.Group A was given powder of Bu-shen herbs[2.5ml].Group C was injected with progesterone[8mg/(kg.d)]and Group D with prolactin(8 IU) twice a day in the pregnant day 1~11.The rats in Group E were normal pregnant rats,and Group F was taken in control model.3.All these rats were killed at the pregnant day 12 to observe the expression of SOCS1,SOCS2,SOCS3 from the deciduas using immunohistochemistry and real time PCR.Use serum to observe the cytokine(IL-4,IL-10,IFN-γ,TNF-a) by ELISA4.Statistical analysis:statistical analysis was performed by SPSS 15.0 software.All data were processed by K-W analysis,LSD.Correlation analysis Significance level is P<0.05Results:1.The pregnant rate and total number of litters in Group D were significantly lower than those in Group A,B,C,E(P<0.01)2.The expression of SOCS1,SOCS2,SOCS3 in Group D were significantly lower than those in Group A,B,C,E(P<0.01) using immunohistochemistry3.The expression of SOCS1,SOCS2,SOCS3 in Group A,B,C,E were higher than those in Group D using Real time PCR.and the SOCS1,SOCS2 expression in Group B,C were significantly higher than in Group D(P<0.05),and the SOCS3 expression in Group A were significantly higher than in Group D(P<0.05).4.The ratio of the Th2/Th1 in Group D was significantly lower than those in Group A,B,C,E(P<0.01) using ELISA.5.Correlation analysis indicate SOCS1,SOCS2,SOCS3 postively correlated with IL-10/TNF-a ratio(P<0.01)Conclusion: 1.The expression of SOCS1,SOCS2,SOCS3 in modified bromocriptine-induced abortion model in rats are lower than those Group A,B,C,it may have connections between the SOCS high expression and immune response from Th1 to Th2 in maternal-fetal interface.2.Bu-shen treatment increase the expression of SOCS,and regulate the self tolerance and maintenance of early pregnancy in maternal-fetal interface. |
