PART 1.STUDY ON THE GROWTH INHIBITION EFFECTS OF DEMETHYLZEYLASTERAL ON DIFFERENT CANCER CELL LINES IN VITROObjective To study the growth inhibition effects of demethylzeylasteral on T-24, SPC-A1,SW620,7860,BEL7404,7901 cell lines.Methods After 6 different cell lines were treated by 0,0.625μM,1.25μM,2.5μM,5μM,10μM,20μM,40μM demethylzeylasteral respectively,cells growth activities were assayed by MTT at 0,24,48 and 72 hours.Results Demthylzeylasteral can suppress the growth of the six different cancer cell lines in time-dependent and dose-dependent manner.T-24 and 7901 were the most sensitive to demthylzeylasteral,SPC-A1 and 7860 were insensitive.Conclusions Demethylzeylasteral can suppress the growth of T-24,SPC-A1,SW620, 7860,BEL7404,7901 cancer cell lines in vitro.PART 2.STUDY ON THE GROWTH INHIBITION EFFECTS OF DEMETHYLZEYLASTERAL ON URINARY SYSTEM CANCER CELL LINES IN VITROObjective To study the growth inhibition and cell cycle phases change effects of demethylzeylasteral on Du-145,PC-3,EJ cancer cell lines.Methods After Du-145,PC-3,EJ cells were treated by 0,0.625μM,1.25μM,2.5μM,5μM,10μM,20μM,40μM demethylzeylasteral respectively,cells growth activities were assayed by MTT at 0,24,48 and 72 hours.After 48 hours,observed the change of the cells' appearance by light microscope.At 24,48 hours,cell cycle phases and the apoptosis were inspested by flowcytometery(FCM).Results Demthylzeylasteral can suppress the growth of Du-145,PC-3,EJ cancer cell lines in time-dependent and dose-dependent manner.Cell cycles were arrested at G0/G1 phase and not found obviously apoptotic peak.Conclusions Demethylzeylasteral can suppress the growth of Du-145,PC-3,EJ cancer cell lines in vitro.The mechanism maybe related to arrest cell cycles at G0/G1 phase.
