This study is mainly on the chemical constituents of Astragalus membranac eus Bunge by means of extraction and separation.Quantitative analysis of main chemical compositions in A.membranaceus had been carried out by HPLC-PDA D-ELSD technique.At the same time,extraction process of astragaloside IV was also discussed.AimAstragalus membranaceus Bunge var.mongolisms(Bunge) Hsiao belongs to the Astragalus genus,Leguminosae family.It is an important traditional Chinese her b,which has been reported to be effective in improving the immune system,the respiratory system,and the circulatory system and helpful in regeneration of th e tissue.Previous studies on chemical constituents of this species included sapon ins,flavonoids,polysaccharides,free amino acids,andβ- sitosterol,of which sap onines and flavonoids are more important.At present time,researches on quality evaluation of A.membranaceus are mainl y focused on fingerprint characteristic features.Quantitative analysis of main che mical compositions in A.membranaceus had not been reported except for astrag alosideⅣ.In order to give a sketch of the active constants in A.membranaceu s.Here we reported Quantitative analysis of main chemical compositions in A. membranaceus.MethodN-BuOH fraction was isolated by Sil.gel,Sephadex LH-20,ODS,active carbon chromatogram.The structures were elucidated through spectrum method.We sel ect 6 major ingredient including AstragalosideⅣ(5),AstragalosideⅡ(6),Meth ylnissolin 3-glucoside(1),3'-hydroxy-5'-methoxyisoflavone-7-O-β-D-glucopyranosi de(3).7-hydroxy-4'-methoxyisoflavone(12),and 7,3'-diohydroxy-5'-methoxyisof lavone(13) as reference substances to found analysis method for determining co ntent of active components by HPLC-PDAD-ELSD technique.ResultsThirteen compounds were identified as astragalosideⅣ(5),astragalosideⅡ(6), Methylnissolin 3-glucoside(1),Isomucronulatol 7-glucoside(2),3'-OH-5'-OCH3-is oflavonoids-7-O-β-D-glucopyranoside(3),4'- OCH3-isoflavonoids -7-O-β-D- gluc opyranoside(4),hexadecanoic acid(8),Daucosterol(9),β-sitosterol(7),adenin e nucleotide(10),disaccharide(11),7-hydroxy-4'-Methoxyisoflavone(12),and 7, 3'-diohydroxy-5'-Methoxyisoflavone(13).The contents difference of 6 main com ponents of Astragaloside between Astragalus membranaceus Bge.var.mongholicus(Bge.) Hsiao and Astragalus membranaceus(Fisch.) Bge.was discussed.It was found that th ere were no significant differences between 6 and 13,but the others were remar kably different.Contents of 5,3 and 1 are higher in Astragalus membranaceus(F isch.) Bge.,and contents of 12 are higher in Astragalus membranaceus Bge.var.mongho licus(Bge.)Hsiao,extraction process of astragalosideⅣwas also deeply discussed here,productive rate is 0.2%.
