Platycodon grandiflorum A.DC.belongs to the Campanulaceae family,and the roots of which,Radix Platycodons,have been used as either a food material or a traditional oriental medicine for diseases such as bronchitis,asthma,pulmonary tuberculosis,hyperlipidemia,diabetes and inflammatory diseases.We always study saponins and pharmacological activities of roots of Platycodon grandiflorum A.DC.But studies about chemical composition of stems and leaves of P.grandiflorum is seldom,pharmacological activities still be restricted at extracts.In order to make further use stems and leaves of P.grandiflorum resources,we studied Saponins and biological activities of stems and leaves of P.grandiflorum.The author employed conventional botanical chemical methods,combining P-HPLC to systematically study the components of stems and leaves of P.grandiflorum,and had the assaying of total flavonoids and total saponins from stems and leaves of P.grandiflorum;the antioxidant activity of stems and leaves of P.grandiflorum was determined by DPPH·and O2—·methods and expectorant effect was determined by phenol red method.Ten compounds were isolated from stems and leaves of P.grandiflorum through the conventional chromatography(macroporous resin,sillica gel column chromatography,sephadex gel to abstract)and HPLC isolation method and six compounds were identified by MS,IR,UV,1H-NMR,13C-NMR,and the six compounds are Platycodin D,Platycodin D3,Polygalacin D2,Deapi-platycodin D,β-sitosterol, JiegengnosideⅥ,respectively.The modern pharmacological research shows that Platycodin as the main active ingredient in Radix platycodonis has broad bioactivities,it plays a decisive role in the efficacy of the medicine.The total saponins were determined in six monthes(May,June,July,August,Sepetember,October),results indicated that the content of total saponin is the highest in June.After blooming,the content of total saponin starts to step down and the content in flower becomes to step up in last phase,but it is lower than spring.In recent years,Radix platycodonis has attracted wid-espread attention for its pharmacological activities in immune regulation,anti-inflammatory,analgesic,antipyretic,expectorent,antilipemic,protective cholesterol-lowering,etc.But,report about antioxidant activity is seldom and there is no report about antioxidant activity of monomer ingredients from stems and leaves of P.grandiflorum.The author studied antioxidant activity of extract and monomer ingredients,results indicated that stems and leaves of P.grandiflorum have antidotal effects.EC50 of n-BuOH layer is 0.407 mg/ml.Antioxidant activity of monomer ingredients from higher to lower is JiegengnosideⅥ,Platycodin D,β-sitosterol,Polygalacin D2,Platycodin D3, Deapi-platycodin D.P.grandiflorum has a variety of pharmacological actions.Clinically,it is widely used in the form of peeled radix and the rest part of it were often abandoned.The thesis made an expectorative effect pharmacology experiment with stems and leaves of P.grandiflorum.The results indicated that the expectorative effect of stems and leaves of P.grandiflorum shows similar pharmacological actions to roots of P.grandiflorum under the same concentration and the expectorative effect went up remarkably with an increasing concentration in certain ranges.It contributes a lot to make full use of the P.grandiflorum and expand the medicinal resources.
