In this paper,We take rotavirus which arose infantile diarrhea as research object in Beijing,to develope a more sensitive and specific diagnostic test for rotavirus by applying real-time fluorescence quantitative RT-PCR technology,and to analyze the pollution characteristics, distribution,seasonal fluctuations and source of the rotavirus.Based on the concentration of rotavirus and by the Beta-Poisson model,the health risk was assessed through sewage treatment plants,surface water and groundwater,which were used as landscape and irrigation.The results were as follows:1,VP7 gene sequence was used as a amplification target to establish a rotavirus quantitative fluorescence quantitative RT-PCR detection methods by SYBR Green I fluorescent dyes as fluorescence signal,and the annealing temperature,reaction time,concentration of polymerase and the preparation of template were evaluated.The results showed that the system had a high degree of sensitivity and specificity,It is a new rotavirus detection technology for clinical samples and environmental samples for their fast and high-volume testing.2,During the period of November 2006 and October 2007,140 samples were collected from sewage treatment plants,and there were 36 positive samples of rotavirus by routine RT-PCR, accounting for 25.71%,and 43 positive samples of rotavirus by real-time fluorescence quantitative RT-PCR,accounting for 30.71%;During the period of September 2006 to August 2007,123 samples were collected from landscape water,and the results showed that 60 positive samples of rotavirus by real-time fluorescence quantitative RT-PCR,accounting for 48.78%,Meanwhile,31 positive samples were detected from 109 landscape waters by routine RT-PCR,accounting for 28.44%.3,Our results showed that rotaviruses were mainly detected in autumn and winter from sewage treatment plants(STPs),landscape water and ground waters samples,which was coincided with the high frequency in children seeking medical care for acute diarrhea in Beijing Children's Hospital caused by rotaviruses.In the three sewage treatment plants,rotavirus were mainly detected from October 2006 to February2007,between September and December in landscape waters,occasionally found in other seasons.4,During sampling period,rotavirus was detected in the three sewage treatment plants,with a higher concentration between3.19E+04copy/L~5.52E+04copy/L,after treatment the concentration of rotavirus reduced to 2.12E+04copy/L~6.62E+04copy/L indicating that rotavirus can not be completely removed by current process,rotavirus can transfer to landscape water from sewage treatment plants,and polluted the source of drinking water.Therefore,improving the existing sewage treatment process to protect human health and environmental safety is highly needed.5,Based on the concentration of rotavirus and through the dose-effect model,the health risk was assessed in sewage treatment plants,Pi was 1.99×10-1~5.00×10-1,Pa was 5.82×10-1~9.36×10-1. we foued that surface water and groundwater which were used as landscape and irrigation put human health and environment at risk on same degree.
