Objective:Alzheimer's disease(AD),occurring at presensiumor senectitude,is a neurodegenerative disease whose main symptom is cognitive handicap(e.g,progressive hypomnesia), accompanied by behavioral and psychological abnormalities.The characteristic clinical manifestations of AD are serious decrease in learning-memory function,decline in ability to analyze and judge,emotional changes,behavioral disorders, and even conscious clouding,which have made some serious effect on the elder's daily life and social activities,and finally reduced their life quality.Therefore,the keystone and breakthrough of AD's research consists in the effective improvement of AD patients' learning-memory ability and their life quality.Acupuncture therapy has the advantages of multi-target,multi-avenue,multi-level,and overall treatment,which just accord with modern medicine's multi-targeted therapy strategies for complex diseases as well. The subject starts with the related signal transduction molecules of learning-memory to make the further explore on the central mechanism of AD's acupuncture treatment,which provides the experimental evidence for AD's clinical treatment. Methods:SD rats were randomly divided into 4 groups: normal group,sham-operation group,model group and electroacupuncture (EA) group.Bilateral hippocampus of rats were one-off injected with condensed Aβ25-35 for preparing the AD rats'model.EA(2Hz,1mA,continuous wave,G6805-ⅡEA therapeutic apparatus) was applied to bilateral "Shenshu" (BL23),bilateral "Neiguan"(PC6) and "Dazhui"(GV14) for 20 min,once daily and six times a week for 2 weeks.Test the changes of each group's learning-memory ability with Morris water maze.Detect the protein expression'schanges of cortex of temporal lobe PKA and CREB in each group by immunohistochemistry. And make a further analysis on the effect of EA's intervention.Result:①In the Morris water maze experiment,compared with the sham group,it was significant that the escape latency of the model group's rats was obviously longer(P<0.01).In comparison with the model group,the escape latency of the EA group's rats is significantly shorter(P<0.05).②The cortex of temporal lobe PKA and CREB protein expression of the model group's rats obviously decreased compared with the sham group(P<0.05).While comparison between the EA group and the model group,the expression of the two protein above mentioned of the EA group had more increase(P<0.05).Conclusions:The rat model with its bilateral hippocampal one-off injected Aβ25-35 successes in simulating the AD's behavior study characteristic.What's more,AD rat's learningmemory ability can get significant improvement by stimulating bilateral "Shenshu"(BL23),bilateral "Neiguan"(PC6) and "Dazhui"(GV14) with EA.The mechanism may be activating the PKA-CREB signaling pathway by enhancing the expression of the cortex of temporal lobe PKA and CREB to keep the normal learning-memory processes of the centrum.
