| Diabetes is one of the major diseases which is a threaten to our human beings'health. Bitter melon and puerarin can remedy diabetes in China's traditional medicine. Consequently the research of bitter melon&puerarin hypoglycemic granules possess significant value. In our work, we did a systerm research on the extracting,and separating of the chemical constituents of the bitter melon, and we also developed a bitter melon&puerarin quality standards of hypoglycemic granule. At the same time, we carried out animal experiments in order to study and research the pharmacological action of the bitter melon&puerarin. The main contents as follow:(1) The different extraction methods about bitter melon were used. We had a preliminary analysis the effect of the solvent, temperature, time, the number of returnees to the effects of extraction. According to optimize the extraction of returning, we determined the best extraction method, that we use alcohol as the solvent and extract three times at 80℃, 8 hours each time. This process give a good example for the extraction of other plants. On this base, two compounds were isolated from the Momordica.(2) We identified Puerarin through TLC. The result showed there was no interference, speckle was obviously and specific. We thought TLC can be used as qualitative identification standards. A quality criteria was founded by UV spectrophotometry and HPLC, in which we can control the quantitation of bitter melon polyose and puerarin. The results showed that the methods have highly sensitive, good reproducible and accurate, which can be used for the assaying of granule components.(3) Through the preliminary test for the hypoglycemic effect of the bitter melon&pueraria hypoglycemic granules, we had a conclusion that the bitter melon&pueraria hypoglycemic granules can reduce hyperglycaemia noticeably for the hyperglycaemia animals by STZ-induced, and have dosage effect. Our preliminary test also provided a test basis for the crowd testing. |
