Objective:Ovarian cancer is one of the most common women's reproductive system cancers.It has been confirmed that ovarian cancer is associated with a variety of proto- oncogenes and tumor suppressor genes variation. such as P53 gene, Ras gene,erbB-2 gene, P16 gene. Among them, the tumor suppressor gene P53 and its family members P73 and P63 are important genes directly related to ovarian cancer.P73 and P63 are P53 family members which found sequentially on a basis of gene homology.There is abnormal expression of P73 gene in tumor tissues. Few mutations, usually as diallele expression; Its splice variant extensively reside in ovarian cancer. P63 gene- targeted areas are associated with ovarian cancer-related gene amplification. But mutation in tumor cells is rarely seen. Many studies have shown that the positive expression rate of P63 in serous cystadenocarcinoma is significantly higher than other types of epithelial ovarian cancer,which is expected to become serous ovarian cancer marker. The abnormal expression of P63 and P73 can promote the occurrence and development of epithelial ovarian neoplasm. However, there is no coverage at home and abroad on the relationship between P63 and P73. This study will explore both in epithelial ovarian neoplasm' expression and their relationship and will provide areference indicators for ovarian cancer's diagnosis and treatment. Meanwhile,detecting P73 and P63 genes in primary epithelial ovarian neoplasm and its relationship with clinicopathological features. So that to explore the efection both in occurrence and development of primary epithelial ovarian neoplasm and the prognosis, clinical value and relevance of them.Methods: 120 cases of ovarian tumor specimens were collected (during 2007-2008).Of these, 25 cases of benign ovarian tumor;18 cases of borderline ovarian epithelial neoplasm; 57 cases of malignant ovarian tumor (9 cases of mucinous cystadenocarcinoma;39 cases of serous cystadenocar- cinoma;9 cases of endometrial carcinoma ).Taking 20 cases of normal ovarian tissue as normal control which resectied during the same period from hysteromyoma tumor resection or surgery for non-neoplastic reasons.All patients were aged from 46 to 66 years old. All tissues were 10% formalin fixed and paraffin- embedded solution.All specimens were confirmed by pathology, neither chemotherapy nor radiotherapy treated before surgery. Using immunohistochemical S-P method for detection.Dealing data with SPSS10.0 package.Analyzing results by x2 test and spearman rank correlation. The results are significant difference when P<0.05.Results:1.P73 expressions lowly in normal ovarian tissue,but the positive expression rate of p73 in ovarian benign, borderline and malignant tumors increased gradually. The difference has statistical significance (p<0.05).P73 protein expression in malignant group significantly higher than the normal group and the benign group And there is significant difference between the benign group and the healthy group (P<0.05).2.In the epithelial ovarian cancer, there is no significant difference of P73 protein expression level among different ages, pathological type and lymph node metastasis.But with the increasing of tissue classification and surgical pathologic stage, the expression level of P73 in poorly differentiated group is significantly higher than high differentiation group.The positive rate of P73 inâ…¢-â…£period is significantly higher thanâ… -â…¡period.So the difference has statistical significance.3.There is no P63 expression in nomal ovarian tissue,But the positive expression rate of P63 in ovarian benign, borderline and malignant tumors increased gradually.P63 protein expression in malignant group is significantly higher than the normal group and the benign group. Borderline group compared with normal group, the difference has statistical significance. But the difference between normal and benign group, borderline and benign group,malignant and borderline group are not obvious.4.In the epithelial ovarian cancer, there is no relation between P63 protein expression level and the age, histological grade, and lymph node metastasis. The positive rate of P63 inâ… -â…¡period is significantly lower thanâ…¢-â…£period.So the difference has statistical significance.5.In the epithelial ovarian cancer,positive correlation exists between P73 and P63 expression. (Rs=0.505,P=0.000).Conclusions:1.In the developing process of transformation from benign to malignant epithelial ovarian tumor, the trend of expression level of P73 is increased and its expression is higher in malignant tumor than in benign and borderline tumor, and the difference is significantly between the nomal and benign group, which showes that the exaltation of expression of P73 emerging in the early stage of formation of the epithelial ovarian cancer. maybe playing a promoting role in transformation and development of some kind from benign tumor to malignant tumor.2.There is some relation between the P73 expression and the tissue differentiation of ovarian cancer.Expression level of P73 increases from high differentiation group to poorly differentiated group ,suggesting that it can be used as a sign of malignant epithelial ovarian cancer and the pathological grade. meaning a poor prognosis.3.P63 protein showes no expression in nomal tissue but a positive expression in epithelial ovarian tumor. In epithelial ovarian cancer,P63 expression increases. which related to the occurrence of ovarian cancer.4.P63 protein expression is closely related to the clinical type, pathological stage in epithelial ovarian cancer, showing that P63 gene's abnormal expression playing important roles in the development of epithelial ovarian cancer.but the mechanism may be different.5.In epithelial ovarian cancer,expression of P73 and P63 existes a positive correlation,which promptes that P73 and P63's over-expression playing a synergies role in the occurrence and development of epithelial ovarian cancer.
