Objective: To study the effects of Polysaccharide L01 from Laminaria in inhibiting the thrombus formation in Rabbit which is established a thrombus model induced by Intravascular Electricity–Stimulus ,thus providing an experimental basis for developing a new drug for antithrombosis.Methods: 25 rabbits were divided into 5 groups randomly as normal control group,NS control group, LMWH control group ,L-L01 treatment group and H-L01 treatment group. Via the same method to practice carotid artery cannulation on every group ,then administration of fluids and drugs from the marginal ear vein of rabbit .H-L01 treatment rabbits and L-L01 treamtment rabbits were injected L01 7.5 mg/kg and 2.5mg/kg respectively. LMWH control rabbits were injected LMWH 1U/kg,and normal control rabbits and NS control rabbits were injected normal sodium 1ml/kg respectively.Five minutes after administration every group rabbits except normal control rabbits were established a thrombus model induced by Intravascular Electricity–Stimulus, then record the obstruction time and test the activated partial thromboplastin time(APTT) , pronthrombin time(PT),thrombin time(TT),and platelet aggregation.The level of thrombin-antithrombin complex(TAT) in blood plasma of rabbits was measured by enzyme-linked Immunosorbent Assay (ELISA). Making manufacture of pathological section and Electron Microscopy of Ultrathin Section bu using the blood vessel where were suffering Electricity–Stimulus to observe ultrastructural organization of endothelia cells .Results:(1)In the animal experiments ,compared with NS control group, H-L01 treatment group and L-L01 treatment group can extend the time for the formation of arterial thrombosis and significantly prolonged APTT, PT, TT and degraded platelet aggregation, and decreased TAT in the blood plasma, with a significant does-dependent manner. (2)After observed by light microscope, we found that the damage of endangium in H-L01 group, L-l01 group and LMWH control group are not as severe as the NS control group. (3) After observed by transmission electron microscope, we found that the damage of endothelial cell in H-L01 group, L-l01 group and LMWH control group are not as severe as the NS control group.Conclusion:Polysaccharide L01 from Laminaria has inhibitory action in the activation of thrombinogen or the activity of thrombin and L01 also has the function of protection of vascular endothelial cell to inhibit the thrombus formation .
