| Aims: To investigate the expression of TKTL1,M2PK,p-Akt and correlation with HPV16?18 in cervical intraepithelial neoplasia (CIN) and cervical carcinoma.Definite the significance of TKTL1,M2PK,p-Akt in judging the prognosis in cervical carcinoma. In order to offer a new strategy to cure the disease.Methods: 130 patients accepted colposcope or operation with pathologic diagnosis were collected randomly at gynecological department of the Second Affiliated Hospital of ShanXi Medical University from January 2006 to March 2007. 20 cases cervical intraepithelial neoplasiaâ… (CINâ… ),30 cases cervicalintraepithelial neoplasiaâ…¡(CINâ…¡), 30 cases cervical intraepithelial neoplasiaâ…¢(CINâ…¢) ,50 cases uterine cervix cancer. And the 20 normal cervix tissues operated because of hysteromyoma were studied as controls. We examined the expression of TKTL1,M2PK,p-Akt by immunohistochemistry. At the same time HPV 16,18 is detected by Fluorescence quantitative Polymerase Chain Reaction (FQ-PCR). 50 patients with uterine cervix cancer were followed up, drawing the survival curve.Result:1.The overall positive rate of TKTL1 was 81.3%, the expressions of TKTL1 were significantly different between two groups arbitrarily besides CINâ…¡andCINâ…¢(P>0.05). The expression rates of TKTL1 was significantly higher following the development of cervical lesions(P<0.05).In addition, there was a positive correlation between TKTL1 and clinical stage in cervical carcinoma.2.The overall positive rate of M2PK was 73.3%, The expressions of M2PK in CINâ… ,CINâ…¡and CINâ…¢were not significantly different(P>0.05) . The expression rates of M2PK was significantly higher following the development of cervical lesions(P<0.05).In addition, there was a positive correlation between M2PK and clinical stage in cervical carcinoma.3.The overall positive rate of p-Akt was 75.7%, the expressions of p-Akt were significantly different between two groups arbitrarily besides control group and CINâ… ((P>0.05)),CINâ…¡andCINâ…¢((P>0.05)). The expression rates of M2PK was significantly higher following the development of cervical lesions(P<0.05).In addition, there was a positive correlation between M2PK and clinical stage in cervical carcinoma.4.The positive detection rate of HPV16,18 increases significantly following the development of cervical lesions(P<0.05). Close positive correlation was observed between the expression of TKTL1﹑M2PK﹑p-Akt and HPV16,18DNA.5.The survival rate was significantly different between TKTL1(+),TKTL1(++)and TKTL1(+++)(P<0.05). The survival rate was significantly different between M2PK(+),M2PK(++)and M2PK(+++)(P<0.05). The survival rate was significantly different between p-Akt(+),p-Akt(++)and p-Akt(+++)(P<0.05).Conclusion: The positive expression rates of TKTL1,M2PK,p-Akt was significantly higher following the development of cervical lesions, and had a positive correlation with clinical stage in cervical carcinoma. They can succeedly react glucose metabolism feature in the cells of uterine cervix cancer. It could be helpful for providing assistance for judging prognosis and treatment by blocking glycolysis. |
