Objectives: To study antitumor activity and mechenism of DCN on carcinomas.Methods: 1.The S180 was transplanted in 24 mice, then they were divided randomly into 3 groups to receive different treatment saline group(pcDNA3.1(+) vector group and pcDNA3.1(+)-DCN group).Then ten days later, execute the mice;2.Tumor weight and body weight were observed. The weight of spleen and thymus were detected respectively, the inhibition ratios of the tumor weight, the spleen index and thymus index were calculated out;3.the function of monovalent phagocyte system, the antipersonnel activity of naturalkiller cells ;and the the function of proliferative response of T/B lymphocyte were measured;4. the percentage of CD4+CD25+T cells in the spleen were detected by flow cytometry. Results: 1.The DCN could obviously restrain the tumour growth,increase the levels of the inhibition ratios of the tumor weight and the spleen index and thymus index in tumor-bearing mice (P<0.05);2.the activity of NK cell, monovalent phagocyte and the function of proliferative response of T/B lymphocyte were signifieantly higher than that other group(P<0.05);3. the proportion of CD4+CD25+regulatory T cells in spleen was significantly lower than control group.Conclusions: DCN could significantly inhibite the growth of tumor cell, and enhance the immunological function of mice bearing S180 in vivo. DCN could depress the secretion of immunosuppressive molecules from S180 tumor cells.
