| Objective To investigate the relationship of plasma homocysteine and the gene polymorphism of methylintetrahydrofolate reductase(MTHFR) with recurrent spontaneous abortion.Methods MTHFRC677T genotype were analyzed by a technique of polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP) in recurrent spontaneous abortion group(60 cases) and control group(60 cases), and the plasma HCY was measured by means of SMT.Result (1) There have no significant differences of genotypes of MTHFR C677T between control and cases, MTHFR 677TT genetype frequency and T allele frepuencies of RSA pregnant women were higher than control group.(2) There have significant differences of MTHFRC677T genotypes of villi between control and cases, MTHFR 677TT genetype frequency and T allele frepuencies of villi were higher than control group.(3) The plasma HCY in RSA cases(13.88±2.37) was significantly higher than in controls(9.77±2.00)(4) There have significant differences of plasma HCY of three genotypes of MTHFR C677T, The plasma level of HCY was markedly higher in the subjects with homozygous allele than that in the subjects with common allele.Conclusion(1) MTHFR 677C→T mutation can affect the levels of plasma HCY.(2)increased plasma HCY levels of morbidity associated with RSA.(3) Embryonic tissues MTHFR C677T villi gene locus mutation than the blood of pregnant women MTHFR C677T gene locus mutation can increase the incidence of RSA. |
