Genetic Association Research Between Mutation In CYP3A4 Gene And Erxian Soup Response

ObjectiveTo investigate the genetic association between mutation in cytochrome P4503A4(CYP3A4) gene and Erxian suop response.To provide a basis for individualized medicine;To identify Erxian Clinical application of the security;detection Erxian suop on plasma luteinizing hormone(LH), follicle-stimulating hormone(FSH),free testosterone(FT) content.MethodsSeventy-six patients with male climacteric syndrome of both Yin and Yang deficiency received a treatment of erxian suop with one month.The severity of symptoms and responses to Erxian suop were quantizated by the method of Department of Psychology in Istanbul Bosphorus University and Clinical Research Guidance Principle on New Drug of Traditional Chinese Medicine published by China.A single nucleotide polymorphism(SNP) at position rs2242480 of CYP3A4 gene were indentified by polymerase chain reaction(PCR) in order to analyse the relationship between Erxian response and CYP3A4 single nucleotide polymorphism.Results1.76 cases of male climacteric syndrome in patients treated by Erxian Decoction,in which efficacy index＞49 cases accounted for 30 percent, accounted for 30 percent of the 27 cases,Erxian suop in treatment of male climacteric syndrome efficient as follows:64.47%.2.the treatment group and the normal control group,genotype and allele distribution between the no significant difference(P＞0.05),that the demographic distribution of the two groups no significant difference.3.Erxian suop treatment group there were 49 cases of the 30 cases for CYP3A4rs2242480 genotype C/T(61.22%),invalid group there were 27 cases of the 17 cases for CYP3A4rs2242480 genotype C/C(62.96%).4.CYP3A4rs2242480 effective treatment of persons of different genotypes before treatment,after treatment of kidney deficiency of yin and yang of two main card,at no significant differences in disease score(P≥0.05).5.CYP3A4rs2242480 effective treatment of persons of different genotypes before and after treatment,physical fitness,cardiovascular,mental psychological symptoms,sexual function had no significant difference in symptom score(P＞0.05).6.the treatment group after taking Erxian suop luteinizing hormone and reduce the plasma concentration of creatinine(P＜0.05),follicle-stimulating hormone,free testosterone,as well as alanine aminotransferase was no significant difference(P＞0.05).Erxian suop taken after the plasma luteinizing hormone to reduce in line with the current research literature, and follicle-stimulating hormone had no significant differences in function of the liver and kidney damage.ConclusionThe patients with genotype of C/T have higher efficiency,while with genotype of C/C have lower efficiency among patients,with CYP3A4 rs2242480 gene when treated with erxian soup.
