Objective:Through retrospective investigation of patients with osteoporosis from 1997 to 2004 in Guangzhou military general hospital,we summarized the theory and clinical practice experience on osteoporosis'prevention and treatment.Then we established the animal model of retinoic acid-induced osteoporosis,and disussed the mechanism of how Bushen Zhuanggu particle prevented and treated osteoporosis.Method:1,To collect tutor'new progresses in theoretical and clinical research on osteoporosis-related bone pain past few years,analyze and summarize the experience on treatment of osteoporosis pain. 2,We chose 3-month-old SD rats for the test,and randomly divided them into 5 groups according to the weight:normal control group,osteoporosis model group,Bushen Zhuanggu granules with high and low dose group,Xianling Gubao positive control group.In the morning all the rats were given retinoic acid for 2 weeks,at the same time in the afternoon with subjects drugs for 3 weeks, and fast in the end.We anesthetized them with 10%Chloral hydrate by intraperitoneal injection,and took the following steps:①got the blood preparations through abdominal aortic bleeding,isolated the blood serum as soon as possible,put them into refrigerator -20℃for preservation,and measured the hydroxyproline/creatinine(HOP/Cr),serum osteocalcin(BGP) levels.②killed them,took both tibia,removed the attachment of soft tissue, fixed,dcalcified,dehydrated,paraffin-embedded,cut into slices,HE stained,measurements and calculate the percentage of trabecular bone area, trabecular thickness,trabecular bone volume and trabecular bone separation with high-resolution color graphic pathological analysis system of 10×10 times light microscope,and observe the upper tibia pathomorphology change.③took both tibia,removed the attachment of soft tissue,fixed them with 10% Formalin,and measured the following index according to the instruction of the YLS-16 small animal bone strength meter:the left femur'maximum load test,the 4th lumbar spine'bone structural strength test and the left tibia'structural strength test.Result:1.According to the chinese medicine practitioners,the osteoporosis pain had the relatinonship to the theory of"stagnant blood"and"blood stasis",based on the traditional pathogenesis,my tutor dicussed osteoporosis from stagnant blood,the difference between"stagnant blood"and"blood stasis"firstly on the pathogenesis,clinical manifestations,treatment, medication,and pointed out the theory of"stagnant blood"played an important part in the osteoporosis,increased the effect of removing blood stasis,and set Calcitonin which was recognized as the main treatment of osteoporosis pain as the positive control group.Finally the effect of that Bushen Zhuanggu particles treated the postmenopausal osteoporosis and osteoporosis pain in older men was affirmed:2.The results of animal experiments①Compared to model control group,in the Bushen Zhuanggu Granules with low-dose,the HOP/Cr value significantly reduced.There was significant difference(P<0.01),Compared to the positive control group,there was significant difference(P<0.01).②Compared to the model control group,in the Bushen Zhuanggu granules group the percentage of bone area and thickness increased,and bone volume and trabecular separation reduced in the proximal tibia cancellous bone trabecular,there was significant difference(P<0.01), Compared to the positive control group,there was no significant difference (P>0.01).③Compared to the control model group,there was no significant difference about the left femur'maximum load,the 4th lumbar'structural strength,the left tibia b maximum load,but indicated a increasing trend, especially in the Bushen Zhuanggu low dose group,the left femur'maximum load,the left tibia'maximum load,the 4th lumbar structural strength increased by 16.2%,17.7%,15.4%.Conclusion:From clinical cases,we know Bushen Zhuanggu granules playthe same effect as Calcitonin in osteoporosis pain,and the experimental results show that the volume and thickness increased in the group of bushen Zhuanggu particles, and the sensitivy indicator of Osteoclast- serum HOP/Cr decreased significantly,so we can infer:Bushen Zhuanggu particles can reduce bone resorption caused by osteoclast,protect the bone morphology,so that it can treat osteoporosis,especially osteoporosis pain.
