Objective To expore the molecular mechanisms in the airway inflammation and mucus hypersecretion induced bu intrateacheal instillation of LPS.To investigate the effects of the extracts of Pinellia(EP) on a rat model of airway mucus hypersecretion by LPS and explore the molecular mechanisms.Methods 30 Wisatr rats were randomly divided into 5 groups(n=6 in each group),Rat airway mucus hypersecretion models were induced by intratracheal instillation of LPS.Rats were orally administered with EP (10g/kg),EP(30 mglkg),or EP(60g/kg) for 4 days.Histopathology examination of lung tissue section stained with HE and AB-PAS.MUC5AC and its mRNA expression were assayed by immunohistochemistry and RT-PCR,the expression of AQP5 mRNA were detected by RT-PCR;ELISA technique was performed to detect TNF-αin bronchoalveolar lavage fluid(BALF).Results(1) LPS significantly induced the overexpression of MUC5AC mRNA in the lung and its protein in bronchial epithelia,and increased the release of mucins,TNF-αin BALF.Moreover LPS decreased the expression of AQP5 mRNA in the lung;(2)EP(60g/kg) significantly decreased the expressions of MUC5ACmRNA in the lung and its protein in bronchial epithelia(p<0.05),EP(10g/kg) and EP(30g/kg) also decreased those(p>0.05);(3)EP(60g/kg) significantly increased the expressions of AQP5mRNA in the lung(p<0.05),EP(10g/kg) and EP(30g/kg) also increased those(p>0.05);(4)EP(60g/kg) significantly decreased the release of TNF-αin BALF(p<0.05),EP(10g/kg) and EP(30g/kg) also decreased those(p>0.05) (5)The up-regulated expression of MUC5ACmRNA was positively correlated with level of TNF-α;AQP5 mRNA was negatively correlated with MUC5ACmRNA and TNF-α.Conclusions Rat airway mucus hypersecretion models were induced by intratracheal instillation of LPS.EP(60g/kg) significantly attenuated bronchial MUC5AC expression and increased the expressions of AQP5.airway mucus hypersecretion may be inhaced by EP through the inhibiting of the release of TNF-αin BALF.
