The Clinical Observation And Experimental Research Of Knee Osteoarthritis Treated By The BuShenHuoXueTongbi Formula

Objective:Knee osteoarthritis(OA) is a common degenerative joint disease that seriously endangers the health of middle-aged and older,history of Chinese medicine accumulated a great deal of experience and got a very good effect in the process of clinical treatment by preventing and treating osteoarthritis.BushenyiHuoxueTongbi recipe(DuhuoJisheng Decoction) is effective in treatment of osteoarthritis.This observation further explores the clinical efficacy and mechanism of BushenyiHuoxueTongbi recipe to knee osteoarthritis in the field of the clinical and the experimental.Also elaborates the influence of BushenyiHuoxueTongbi recipe to bone marrow-derived mesenchymal stem cells(BMSC) differentiation,and further elaborates the theory of "Lord of renal bone" which as a base on promoting further clinical applications of BushenyiHuoxueTongbi recipe.Method:In the clinical observation,a total of 40 patients with early and middle stage of osteoarthritis of the knee were randomly assigned to receive Bushenhuoxuetongbi recipe and Celibrex.Outcomes were assessed at multiple points including symptom,walking ability,anf daily life function.In the experimental observation,the Sdmouse BMSC were cultured in vitro and were added with DMEM. After then,Bushenhuoxuetongbi groupwas added with 40ng/ml Dexamethasone,VitC20ug/ml and various concentration of Bushenyiqihuoxue recipe serum and various concentration of blank serum.14 days later,chondrocytes were detected by Toluidine blue staining and Immunohistochemistry.Result:In the clinical observation,both Bushenhuoxuetongbi recipe and Celibrex can decrease the pain,improve the function of the knee,the positive effect rate is 85%and 90%.In the experimental observation,various concentration of Bushenyiqihuoxue recipe serum could induce BMSC differentiate into cartilage cells.Conclusion:For the early and middle stage of osteoarthritis, Bushenhuoxuetongbi recipe can improve the function of the knee,delay the development of cartilage degeneration with little side effect,and significant advantage of economic-effect relationship.And Bushenhuoxuetongbi recipecan could induce BMSC differentiate into cartilage cells,which may be one of its therapeutic mechanisms in osteoarthritis treatment.
