Objective:The aim of this research is to study the differentiation function of cultured preadipocyte.By detecting two indicators,PPARγ2 and PPARγ2mRNA, we want to explore the mechanism of action of Jianpi HuatanFa by regulating the differentiation of fat cell.Methods:In the experiment,we use JianpiHuatanFang,JianpiFang,HuatanFang and Retinoic Acid to feed the rats.Meanwhile,we established blank control group.When the drug concentration in the rats serum is stable,we draw the blood and extract the serum of all the rats in each group to culture the cells.Then we detecting two indicators,PPARγ2 and PPARyγ2mRNA.The data are analysed in statistics.Results:1.The Fourth Day,10%,the quantity of PPARγ2mRNA:Retinoic Acid>Huatan>Blank Control>Jianpi>JianpiHuataniFang(P<0.05);2.The Eighth Day,Huatan,the quantity of PPARγ2mRNA:10%Group>15% Group>5%Group(P<0.05);3.The Eighth Day,JianpiHuatan,the quantity of PPARγ2mRNA:10%Group>5%Group(P<0.05);4.The Fourth Day,Retinoic Acid Group,the quantity of PPARγ2mRNA:10% Group>15%Group>5%Group(P<0.05);5.10%,Huatan Group,the quantity of PPARγ2mRNA:The Fourth Day>The Eighth Day>The Twelfth Day(P<0.05);6.10%,Retinoic Acid Group,the quantity of PPARγ2mRNA:The Fourth Day>The Eighth Day>The Twelfth Day(P<0.05);7.15%,Jianpi Group,the quantity of PPARγ2mRNA:The Fourth Day>The Twelfth Day>The Eighth Day(P<0.05);8.The amount of PPARγ2 in JianpiHuatan Group,Jianpi Group,Huatan Group is significantly lower than Retinoic Acid Group,Blank Control Group.Among all these groups,the JianpiHuatan Group produce the lowest amount of PPARγ2;9.There is no siginifieant difference between Jianpi Group and Huatan Group。Conclusion:Jianpi HuatanFa can decrease the secretion of the expression amount of PPARγ2 and PPARγ2mRNA,and in this aspect,JianpiHuatanFa does a better job than JianpiFa and HuatanFa.The experiment shows us that the spleen- weak and phlegm-dampness is the main etiology and pathology of the abnormity of the matabolism of the fat in Traditional Chinese Medicine.And we can find out part of the mechanism of the treatment of JianpiHuatanFa.So this study have significant importance on preventing and curing the abnormity of the the matabolism of fat by Traditional Chinese Medicine.
