Object:To investigate the expression of KLF5,Sp1 and CyclinDl in human stomach normal mucosa,atypical hyperplasia and malignant tumor,furthermore to study the relationship and clinic significance among the expression ofKLF5,Sp1 and Cyclin D1 in human stomach carcinoma,and the dependability among them,which can provid important theoretical evidence for blockade metastasize,diagnosis and therapy of stomach carcinoma.To explore the role of KLF5,Sp1 and Cyclin D1 on the development and progression of gastric carcinoma.Materials and Methods:78 cases of stomach carcinoma patients,20 case o f atypical hyperplasia tissue and 20 case of normal stomach who underwent su rgical resection and endoscopic biopsy specimens between 2005 and 2008 at th e Second Hospital Attached To KunMing Medical college(20 patients with nor real stomach were come from this period for visit of patients by endoscopic bi opsy and histopathological study confirmed the infiltration of non-cancer cells o r precancerous lesions.Expression of KLF5,Sp1 and CyclinDlwere examined by S-P immunohistochemistry assays.The relationship between expression of K LF5,Sp1 and CyclinD1 in human stomach carcinoma were analyzed by SPSS 11.5 statistics software.Results:1 The expression of KLF5 were obviously lower in the carcinoma group than in the atypical hyperplasia group and normal group,and gastric cancer and normal group KLF5 expression positive rate of significant differences(P<0.01),(respectively P < 0.01).There is no different between the expression of KLF5 in atypical hyperplasia group and normal group(respectively P > 0.05).2 The expression of Sp1 and CyclinD1 were obviously higher in the carcinoma group than in the atypical hyperplasia group and normal group.and gastric cancer and normal group Sp1 and CyclinD1 expression positive rate of significant differences(P<0.01),(respectively P<0.01).There is no different between the expression of Sp1 and CyclinDlin atypical hyperplasia group and normal group (respectivelyP >0.05).3 The expression of KLF5 in stomach carcinoma were obviously lower in the low differentiation group than those of in the midstand high differentiation group (respectively P<0.05).There is no different between the expression of KLF5 in the high differentiation group and in the midst group(respectively P>0.05).The expression of KLF5 in stomach carcinoma were obviously lower inserous tunic tresised group than those of in no serous tunic tresised group(respectively P<0.05). The expression of KLF5 in stomach carcinoma were obviously lower in lymphatic node metastasis group than those of in no lymphatic node metastasisgroup (respectively P<0.05).The expression of KLF5 in stomach carcinoma were obviously lower inâ…¢,W group than those of inâ… ,â…¡group(respectively P<0.01).There is no different between the expression of KLF5 inâ…¢and W group,â… andâ…¡group(respectively P>0.05).The expression of KLF5 in stomach carcinoma were obviously lower in the diameter> 5cm group than those of in the diameter <5cm Group(respectively P<0.05),The expression of KLF5 have no relationship with sex, age,location,histological type and whether or not between the history of Hp infection (respectively P>0.05).4 The expression of Sp1 and Cyclin D1 in stomach carcinoma were obviously higher in the low differentiation group than those of in the midst and high differentiation group(respectively P<0.05).There is no different between the expression of Sp1 and CyclinD1 in the high differentiation group and in the midst group(respectively P>0.05).The expression of Sp1 and CyclinD1 in stomach carcinoma were obviously higher in serous tunic tresised group than those of in no serous tunic tresised group(respectively P<0.05).The expression of Sp1 and CyclinD1 in stomach carcinoma were obviously higher in lymphatic node metastasis group than those of in no lymphatic node metastasis group(respectively p0<.05).The expression of Spl and CyclinD1 in stomach carcinoma were obviously higher in â…¢,W group than those of inâ… ,â…¡group(respectively P<0.05).There is no different between the expression of Sp1 and CyclinD1 inâ…¢and W group,â… andâ…¡group(respectively P>0.05).The expression of Sp1 and CyclinD1 have no relationship with sex,age,size,location,histological type and whether or not between the history of Hp infection(respectively P>0.05).5 The expression of KLF5,Sp1 and Cyclin D1 have relationship with each other (respectively P<0.05).Conclusion:1.The expression of KLF5 were obviously lower in the carcinoma group than in the atypical hyperplasia group and normal group.The expression of KLF5 have relationship with the degree of differentiation,the depth of infiltration,lymphatic node metastasis,size and TNM period of stomach malignant tumour.2.The expression of Sp1 and Cyclin D1 were obviously higher in the carcinoma group than in the atypical hyperplasia group and nonnal group.The expression of Sp1 and Cyclin D1 have relationship with the degree of differentiation,the depth of infiltration, lymphatic node metastasis and TNM period of stomach malignant turnout.3.There was positive correlation between the expression of Sp1 and CyclinD1 in the gastric carcinoma tissues.There was negative correlation between the expression of KLF5 and CyclinD1 in the gastric carcinoma tissues.There was negative correlation between the expression of KLF5 and Sp1 in the gastric carcinoma tissues.4.The lower expression of KLF5 and the higher expression of Sp1 and Cyclin D1 in gastric cancer group are involved in the carcinogenesis and development of gastric cancer,but each of them might play a different role.To monitoring the expression of KLF5,Sp1 and Cyclin D1 in human stomach precancerous lesions,will help to screen out high-risk patients with gastric cancer and provide important theoretical basis with early clinical diagnosis,prognosis and targeted therapy of gastric cancer in the future.
