Objective:To reveal the relationship between female lung cancer in Xuanwei Country and 8-OHdG produced by the DNA(Deoxyribonucleic acid) oxidation which caused by the burning of large amount of bituminous coal.We research the expression of 8-OHdG(8-hydroxy-2-deoxyguanosine) in urine,lung cancer tissues,adjacent cancer tissues and normal tissues of female lung cancer patients who live in Xuanwei Country.Method:The experiment of measuring 8-OHdG in urine methodExperimental group:18 cases of the Xuanwei female lung cancer patients.Control group 1:16 cases of the Xuanwei female benign pulmonary diseases patients.2:10 cases of the Xuanwei male lung cancer patients.3:10 cases of the Xuanwei male benign pulmonary diseases patients.4:17 cases of the Non-Xuanwei female lung cancer patients.5:13 cases of the Non-Xuanwei female benign pulmonary diseases patients.collected early morning urine of The patients,8-OHdG Content in urine was measured using the 8-OHdG ELISA(enzymelinked immunosorbent assay).The urine creatinine was measured using picric acid.The final results of 8-OHdG content was expressed with ng/ mg creatinine.The experiment of measuring 8-OHdG in tissue methodExperimental group:18 cases of cancer tissue,adjacent cancer tissue and normal tissue of the Xuanwei female lung cancer patients.Control group 1:16 cases of benign lesion tissues and normal tissue of the Xuanwei female benign pulmonary diseases patients. 2:10 cases of cancer tissue,adjacent cancer tissue and normal tissue of the Xuanwei male lung cancer patients.3:10 cases of benign lesion tissues and normal tissue of the Xuanwei male benign pulmonary diseases patients.4:17 cases of cancer tissue, adjacent cancer and tissue normal tissue of the Non-Xuanwei female lung cancer patients.5:13 cases of benign lesion tissues and normal tissue of the Non-Xuanwei female benign pulmonary diseases patients.The DNA in lung tissue was extracted out after digestion,and then the 8-OHdG and dG were cut from the DNA chain.The 8-OHdG and dG content were detected using the HPLC(High Performance Liquid Chromatography).The final results of 8-OHdG content was expressed with 8-OHdG/10~6dG..The SPSS16.0 statistical software was applied to statistical analyze the result datas.results:The experiment of measuring 8-OHdG in urine results1.8-OHdG were expressed in the urine samples of all patients,the content of 8-OHdG in urine of lung cancer patients is much higher than that of benign pulmonary diseases patients(P <0.01).2.The content of 8-OHdG in urine of Xuanwei female lung cancer patients is higher than that of Xuanwei male lung cancer patients and Non-Xuanwei female lung cancer patients(P <0.01).3.The content of 8-OHdG in urine of Xuanwei female benign pulmonary diseases patients is much higher than that of Xuanwei male benign pulmonary diseases patients and Non-Xuanwei female benign pulmonary diseases patients.(P <0.01 ).The experiment of measuring 8-OHdG in tissue results1.The content of 8-OHdG reduced gradually from lung cancer tissues,adjacent tissues to normal lung tissue of lung cancer patients and the resalts has statistical significance(P <0.01).2.The content of 8-OHdG in the cancer tissues of cancer patients is higher than benign lesion tissues and normal tissues that of benign pulmonary diseases patients.3.The content of 8-OHdG in the cancer tissues,adjacent tissues,normal tissues of Xuanwei female cancer patients is not higher than that in Xuanwei male cancer patients and Non-Xuanwei cancer patients(P>0.05).4.The content of 8-OHdG in the normal tissues of Xuanwei female benign pulmonary diseases patients is higher than that in Non-Xuanwei female benign pulmonary diseases patients and Xuanwei male benign pulmonary diseases patients(P<0.05).Conclusion:1.8-OHdG have consistency to lung cancer tissues and lung cancer cells in the position.The 8-OHdG produced by DNA oxidative damage may be plays an important role in the occurrence and development of lung cancer.2,The oxidative damage which Xuanwei female suffered higher than the Xuanwei male and Non-Xuanwei female.This may be one risk factor of high incidence of Xuanwei female lung cancer.
