| Objective:To observe the charicteristics of subacute methamphetamine hydrochloride in the poisoning rats and study the function of Caspase-3 and Bcl-2 protein in the methamphetamine mechanism.Methods:Forty male Sprague-Dawley rats were randomly divided into control group (n=10) and experimental group(dividing it into three group after the last injection, n=10.) Rats in the experimental group were intraperitoneal injected with methamphetamine hydrochloride(20mg/kg),and those in the control group were injected with saline with the same volume(twice a day at 8:00 and 20:00 respectively for 4 continous days),then,observing their behavioral changes.Examining the protein expression of Caspase-3 and Bcl-2 of poisoning rats in related encephalic region by immunohistochemistry technique and analyzing the test results through image.Results:Both Caspase-3 and Bcl-2 are featured with the similar expression changes in the different areas of brain with obviously masculine signals.At the first day,Bcl-2 chalks peak after the last rejection,then gradually decreases,so does Caspase-3 at the fourth day.Conclusion:1.Setting up the model of subacute poisoning of methamphetamine.2.The abnormal expression of Caspase-3 of rats Sub-acute poisoning of methamphetamine in PFC, Cpu,CA,shows that it takes part in the Methamphetamine-Induced Neurotoxicity.3. The abnormal expression of Bcl-2 of rats Sub-acute poisoning of methamphetamine in PFC,Cpu,CA shows its close connection with protection of Methamphetamine-Induced Neurotoxicity. |
