[Objective]To establish the epilepsy model in rabbits with penicillin cerebral cortex injection,and to do the rearch of the impact of hypothermy physiological saline lavage to Cortex in epilepsy rabbits.To find an original and utility method to deal with subsequent epilepsy when making the anaesthesia arousal in the surgery of the cerebral functional field[Methods]In the first part of the experimen,we divided 24 rabbbits randomly into 4 groups with 6 rabbits in each one,the 0℃Group,4℃Group,8℃group, and normal temperature group,and establish the epileptic rabbits model by injecting the penicillin into the cortex.After that we make the lavage with 0℃,4℃,8℃,and normal temperature physiological saline respectivly into the 4groups rabbits.To confirm the effect of the epilepsy control,the lavage were lasted 5mins after the seizures stoped.We compare the behaviors and the ECoG of the rabbits in each groups, focus on recording the time span of the epilesy control,pick up the slice of the rabbit's cortex 8 hours after the seizuress were well controled for pathology test which is aim at comparing the impact of the different temperature in each groups,and select the most effective temperature in seizures control.In the second part of the experiment,60 rabbits were randomly divided into selected temperature part(trail group) and normal temperature part(control group 25℃).In each part,the rabbits were divided in to 5 groups,the seizures group,5mins, 2hours group,6hours group,and 12hours gourp based on the observation time point. (totally 10 group,n=6).As the model been made in the first experiment part,we lavage the cortex with the selected temperature physiological saline,lasting the lavage 5mins after the seizuress stoped to confirm the effect of the epilepsy control.Then we take the specimens of the bilateral hippocamps on the 5 observation time points,and test them by the high-performance liquid chromatography apparatus to know the quantity of GLU,Asp,GABA,Gly.After that,we compare the value Glu/GABA,and analyse the mechanism of well controlling the epilepsy by lavaging the cortex with low temperature physiological saline.[Results]In the first part of the experiment,the modelings were made in the every groups.The time span of the seizures in each groups are 0℃group:264±22s,4℃group:289±25s,8℃group:428±18s,normal temperature group:904±21s,these results were consistent with the behaviors and EcoG performance.The time span of the seizures in low temperature groups were significantly less than the normal temperature group.These were statistically significant differences(p<0.01).The 0℃group and 4℃Group time span were less than 8℃group,a significant difference(p<0.05).The pathology test showed some injured tissues in 0℃group with no apparent damage in 4℃group and 8℃group.So 4℃group was selected as the most suitable temperature in seizures control.In the second part of the experiment,we set 4℃group as the experimental group, and the normal temperature group as the control group.Result:(1) After the seizures were controlled,in 4℃group,the quantity of Glu on different observation time points 5mins,2hours,6hours are significantly lower than normal temerature group(P<0.01). On 12hours observation time point,Glu quantity is lower than the normal temperature group(P<0.05).After the attack(Inject the penicillin) and controll of the seizures,the Glu quantity drops obviously compared with the moment when seizures are happening on every observation time points in the 4℃group,especially on the time point of 6hours.(2)The quantity of GABA in 4℃group was higher than the normal temperature group on the obervation time point 12hours(P<0.01),with no apparent difference in other time points(P.>0.05).On the obervation time point 6hours,the quantity of GABA elevates continuously in both groups,with a light drop in the 4℃group and significant drop in the normal temperature group during 6-12hours of the observation time point.(3)The content of the Asp in 4℃group were lower than the normal temperature group on every obervation time points(P <0.05),with a durative drop 5mins after the seizures stoped in both groups.(4)The quantity of the Gly in 4℃group was lower than the normal temperature group in the observation time point 2hours,6hours and 12hours(P<0.05),with an increace after the seizure period and on every time points compared with it's quantity before seizures happened.The Gly droped 5mins after the seizured stoped in 4℃group and 2hours after the seizured stoped in normal temperature group.(5) The Glu/GABA ratio in 4℃group was lower than the normal termperature group in every observation time points(P<0.05),and always kept in a low level.[Conclusions]The cortex lavage by the hypothermy physiological saline can effectively control the epileptic seizures caused by the injection of the penicillin in rabbits.The machinism of it is to well regulate the balance of EAA/IAA,with the most effective temperature at about 4℃.
