Objective:To analyze the infection characteristics of HPV genotypes and the significance of multiple HPV infection as well as the amplification of hTERC gene of cervical lesions in Shenzhen area,in order to provide an objective basis for the prognosis of cervical lesions and the screeing for low or high lesions of cervix.Methods:(1) Extraction of DNA from specimens of cervical cytology and to detect the genotyping of HPV in cervical lesions by low density gene chips.To analyze the popular distribution of HPV gene subtype and explore the relationship between the multiple HPV Infection and cervical lesions.(2) Extraction of cervical exfoliated cells from the residual specimens of liquid-based cytology and preparation of monolayer cells slide.To study the amplification of gene hTERC in cervical lesions by FISH and explore the relationship between the amplification of gene hTERC of the CIN or cervical cancer.Results:(1) In 287 samples,80.8%of those were checked out HPV and the major epidemic subtypes of HPV were HPV16(44.3%),31(10.3%),51(9.7%),52(13.4%),58(16.4%).(2) The multiple infection rate of HPV in CINⅠ,CINⅡ,CINⅢand cervical squamous cancer were 45.5%,42.1%,33.3%and 26.9%.There was no statistics difference of HPV multiple infection between the adjacent groups,however,statistics difference was noted between the groups of CINⅠand CINⅢ(P<0.05);the significant difference was found between the groups of CINⅠand cervical squamous carcinoma(P<0.01).(3) There was an age central tendency in multiple HPV infection and the rate of the multiple HPV infection in 19-40 and 41-71 age groups were 44.21%and 30.6%.The difference of multiple HPV infection between the 19-40 age group and 41-71 age group had a statistical significance(P<0.05).(4) The amplification rate of gene hTERC in CINⅠ,CINⅡ,CINⅢand cervical cancer groups were 11.1%,54.5%,71.4%and 81.8%.There were no gene amplification in the 7 patients of cervicitis group.Statistics difference of gene amplification in hTERC was found between Low grade cervical lesions(cervicitis and CINⅠ)and high grade cervical lesions(CINⅡ-Ⅲand cervical carcinoma)(P<0.01).(5) The forms of gene amplification in hTERC were major in diploid,tetraploid and aneuploid. There were no parallel relationships between the groups of CIN and cervical cancer in the HPV infection and the gene amplification in hTERC. Conclusion:1,In Shenzhen area,the major epidemic subtypes of HPV were HPV16(44.3%), 31(10.3%),51(9.7%),52(13.4%),58(16.4%).2,The multiple HPV infection is not an indicator of severity of cervical lesions.May be the severity of cervical lesions did not rely on the quantity of the HPV multiple infection but depend on the virulence of HPV genetype.3,The amplification in gene hTERC wes a useful auxiliary indicators for the screening of high grade cervical lesions and maybe indicate the progress of cervical lesions.
