Objective: To investigate the proteomics change of synovial fluid in knee joint with osteoarthritis. Seek for an early diagnosis and monitoring of knee osteoarthritis biomarkers.Methods: In accordance with the principle of random selection of patients with knee osteoarthritis, in patients with osteoarthritis are used on the American Rheumatism Association of knee osteoarthritis to select diagnostic criteria for diagnosis. The selected patients with OA were divided into early, middle and late stages through the Kellgren and Lawrecne five classification scheme of radiology diagnostic criteria. Randomly selected early, middle and late stage of the 10 patients with a total of 30 patients as the experimental group. Location close to the same period 10 cases of knee joint synovial fluid as a healthy control group. Taking the knee joint fluid of the experimental group and control group, Clinprot technology platform used, the use of matrix-assisted laser desorption/ionization time of flight mass spectrometry (MALDI-TOF-MS) for synovial fluid analysis of the protein group, to map out the protein spectra to identify differences in protein expression.Results: Through MALDI-TOF-MS analysis, we found two significantly different protein peaks appears at m/z1864.28 and m/z5225.63 in the OA group. There is two significantly different protein peaks appears at m/z1864.33 and m/z1448.15 between medium and early,late groups.Conclusion :Two significantly different protein peaks emerged in the experimental group compared with the control group, consider two kinds or 2 family protein; Statistically significant difference. It shows that the proteins content in synovial fluid of patient's knee joint was higher than healthy people, there are potential diagnostic significance.
