Objective: Diabetes Mellitus is chronic metabolic disorders.diseases of which have the highest ortality and incidence in the world.There are two forms of diabetes:type 1,insulin-dependent diabetes mellitus,whose pancreaticβ-cell are destroyed by an autoimmune reaction,leading to a complete lack of insulin.Type 2,non-insulin-dependent diabetes mellitus,which is caused by multi-factors including pancreaticβ-cell defect,tissue sensitivity reduction,and insulin resistance.95% of the diabetes are type 2 diabetes mellitus.Type 2 diabetes mellitus due to lack of insulin and insulin resistance, hyperglycemia and hyperlipemia metabolic disorders,hyperlipemia can influence blood viscosity,thus cause hemorheological change. Daphnetin has pharmacological effects of reducing thrombosis and blood lipid.Diabetes Mellitus has become a focus of modern medicineMethods: Wister rats were randomly assigned into two groups:normal control groupand diabetic group.Normal control group rats were fed with the normal diet, while diabetic group were fed with the diet containing 10%sucrose, 15%lard, 2%cholesterol and 0.25%cholic acid.Hyperglycemia was developed by intrapenitoneal injection STZ (30mg·kg-1) in diabetic group rats after 12 weeks on this diet to set up rats model of type 2 diabetic.Diabetic rats were randomly devided into seven groups: diabetic model group,xiaoke pills group 3.0g/kg,daphnetin group 0.5g/kg,daphnetin derivatives high dose1.0g/kg,middle dose 0.5g/kg,low dose 0.25g/kg,union medication administration group 3.5g/kg.11 rats evrery group.13 weeks,xiaoke pills group,daphnetin group,daphnetin derivatives high dose,middle dose,low dose,union medication administration group i.g administration during 4 weeks.one time a day,volume of 10ml/kg.normal group and diabetic group fed with distilled water one time a day for 4 weeks.All animals were assayed the values of the rate of platelet aggregation,whole blood viscosity,thrombosis,blood flow velocity,vascular diameter after four weeks .Results: 1.Compared with normal control group, blood platelet maximum agglutination obviously enlarge(P<0.001) , xiaoke pills group, daphnetin group(3.0g/kg,0.5g/kg )and union medication administration group 3.5g/kg can inhibit ADP induced type 2 diabetic mellitus rats blood platelet agglutination(P<0.01 or P<0.01).2.compared with normal control group, whole blood viscosity of model obviously increased(P<0.001), Compared with model group,xiaoke pills group, daphnetin group and daphnetin derivatives group high dose,middle dose(3.0g/kg,0.5g/kg ,1.0g/kg,0.5g/kg)and union medication administration group 3.5g/kg can decrease type 2 diabetic mellitus rats whole blood viscosity(P<0.01 or P<0.05, or P<0.001).3.Compared with normal control group, thrombus longitude of model group obviously growth (P<0.01),Compared with model group, xiaoke pills group, daphnetin group and daphnetin derivatives group high dose,middle dose(3.0g/kg,0.5g/kg ,1.0g/kg,0.5g/kg)and union medication administration group 3.5g/kg can inhibit thrombogenesis of rats.(P<0.05 or P<0.01).Compared with normal control group,thrombus weight of model group obviously growth (P<0.001),Compared with model group, xiaoke pills group, daphnetin group and daphnetin derivatives group high dose,middle dose(3.0g/kg,0.5g/kg ,1.0g/kg,0.5g/kg)and union medication administration group 3.5g/kg can inhibit thrombogenesis of rats.(P<0.05 or P<0.01).4.Compared with normal control group,model group blood flow rate obviously slow-moving(P<0.01).Compared with model group,xiaoke pills group,daphnetin group and daphnetin derivatives group high dose,middle dose(3.0g/kg,0.5g/kg,1.0g/kg,0.5g/kg)and union medication administration group 3.5g/kg can improve diabetes mellitus rats blood flow rate(P<0.05,or P<0.01).Compared with normal control group,vascular diameter of model group obviously narrowing(P<0.01),Compared with model group,xiaoke pills group, daphnetin group and daphnetin derivatives group high dose,middle dose(3.0g/kg,0.5g/kg,1.0g/kg,0.5g/kg)and union medication administration group 3.5g/kg can enlarge diabetes mellitus microvessel diameter (P<0.05).conclusion: daphnetin and daphnetin derivatives can inhibit type 2 diabetic mellitus rats blood platelet maximum agglutination,decrease whole blood viscosiy,inhibit thrombogenesis, improve diabetes mellitus rats blood flow rate and enlarge microvessel diameter.
