F-2 toxin(Zearalenone,ZEA),a kind of estrogen produced by sickle germ,has great reproduction toxicity and teratogenic action.Meanwhile,it can reduce the feedstuff of livestock,resulted in growth reduction,immune suppression, multiplication drawback,cut down the quality of poultry and livestock production, and bring a great economy loss of animal husbandry.F-2 toxin has become the secondary killer in raising hogs or pigs,and even threatened people health.For all of the above,the research was carried on.Research purpose:discussing the study of F-2 intoxation safety action of tea polyphenols and gold theragran on male mouse,comparing the restrained toxin action of two medicine,interpreting controlling toxin mechanism.In order to control toxicity in vivo,supply scientific theory foundation for controlling preparation.Research methods:30-old mouse of 80 was divided into 8 groups randomly, (Ⅰ) control group,(Ⅱ) F-2 toxin group,(Ⅲ-Ⅴ) F-2 toxin+Tea Polyphenols group(including high,middle,and low concentration),(Ⅵ-Ⅷ) F-2 toxin+Gold Theragran group(including high,middle and low concentration) respectively.Every morning,the mouse inⅡ-Ⅷgroup was injected 10μg/0.5ml F-2 toixn,the mouse inⅠgroup was injected 10%alcohol 0.5ml in each one by intragastric administration,in the afternoon,the mouse inⅠ,Ⅱgroup were injected 0.5ml distilled water,Tea Polyphenols water solution of corresponding concentration 0.5ml in each one ofⅤgroup,Gold Theragran extractive water solution of corresponding concentration 0.5ml in each one ofⅥ-Ⅷgroup by intragastric administration.Didymus weight was weighed after completing the experiment,the change of body weight,didymus quality,and didymus organ module were compared.morphology change and apoptosis of didymuscell granule were detected by pathohistology and TUNEL respectively,SOD and MDA change were detected by method routine,expression of P53, Bcl-2and Bax was deteced by RT-PCR method.Research results:comparing with control group,tea polyphenols group and gold theragran group,weight and organ module of mouse didymus degraded obviously in F-2 toxin group,while body weight has insignigicant change.Cell granule apoptosis have results by TUNEL,tea polyphenols and gold theragran have some safety action on didymus apoptosis which was resulted by F-2 toxin,and apoptosis rate has the tendency of degression with the dosage augment.SOD activity and MDA content results:SOD activity degraded obviously in F-2 toxin group,each dosage group of tea polyphenols and gold theragran has the most significant deviation compared with F-2 toxin group(P<0.01),and SOD activity has the tendency of upgrade compared with the dosage of tea polyphenols and gold theragran augmentation,MDA content raised obviously in F-2 group,other groups have the most significant deviation compared with F-2 toxin group(P<0.01).MDA content tapered with the dosage augmentation.The results of the expression of P53, Bcl-2 and Bax in didymus tissue by RT-PCR:each gene protein expression in control group,the P53,Bax are all higher than control group,and Bcl-2 is obvious lower than control group.After using tea polyphenols and gold theragran,the expression of P53,Bax have obviously degrade with F-2 toxin group in each dosage group,the expression of Bcl-2 has step up,all show significant deviation,also present docs-effect relation.Research conclusion:①F-2 toxin can encourage didymus apoptosis of male mouse,tea polyphenols and gold theragran have some depressant effect on apoptosis which was resulted by F-2 toxin.②F-2 toxin can make SOD activity reduce and MDA content increase,Chinese herbal medicine tea polyphenols and gold theragran can inhibit the strong oxidization of F-2toxin on organism,strengthen SOD activity and reduce MDA content in didymus tissue.③P53,Bax and Bcl-2 participate in regulating the didymus apoptosis of male mouse,F-2 toxin can strengthen the expression of P53 and Bax,inhibit Bcl-2 expression,tea polyphenols and gold theragran can strengthen Bcl-2 expression validly,and reduce P53 Bax protein expression.④the reason of the controlling effect of tea polyphenols and gold theragran on F-2 toixn is because the strong antioxidation and to influence the expression of related gene.
