ObjectivesBone defects were common conditions caused by trauma,sarcoma or infection.Small bone defects can be repaired automatically by bone induction,conduction or regeneration.Large bone defects will not heal without bone transplantation.The current bone defects repairing materials include:Autogenous bone,allogenic bone,artificial materials,composite materiel etc.All of these materials were not ideal.It is clinical desirable to find ideal materials to repair larger bone defects.Bone tissue engineering may provide an alternative for bone defects repairing.The aim of this study was to observe the reaction of local and whole body and the healing time with BME-10X hydroxyapatite/collagen as a bone graft substitute for repair of bone defects,and evaluate the clinical effects of BME-10X hydroxyapatite/collagen.MethodsThirty patients including 12 males and 18 females aged 17-78 years with averaged 58 years,admitted to Department of Orthopedics,Tianjin Tianhe hospital were selected between Febrauy 2007 and December 2007.As the control group,28 patients including 14 males and 14 females aged from 20 to 81 years old that averaged 52 years old didn't receive BME-10X hydroxyapatite/collagen.There were 24 cases with bone defects caused by crushed fracture,the BME-10X hydroxyapatite/collagen was implanted in the defects after surgical procedure and internal fixtion;there were 3 cases with nonunion of fractures,the BME-10X hydroxyapatite/collagen was implanted after excising the scar and ossified tissue in fracture and reaming the marrow cavity;there were 3 cases with bone tumor-like defects,BME-10X HAC was implanted in defects after excising the tumor-like tissue.The implantation amount was averaged from 0.4g to 4.0g.Clinical and X-ray photograph assessments were performed consecutively during the follow-up of 6 months.ResultsAll the patients were followed up from 2 to 10 months with the average of 6.3 months.All of the incisions in 30 cases obtained the primary healing.No fever was found in the 30 patients during follow-up,and there was no adverse reaction in the implanted site of BME-10X HAC.Results of X-ray after implantation of BME-10X HAC:There were vague bound between the implanted site and bone tissue around defects 1 to 3 months after operation;from 3 to 6 months after operation,new bone was found in implanted parts,and the graft material was fused with bone tissue,the bone density was near to normal tissue,the defects were almost repaired.Two patients in control group were nonunion of fracture.All the patients that received implantation of BME-10X HAC obtained clinical healing of bone with the average time of 3.8 months. ConclusionBME-10X HAC has good biocompatibility and biodegradability;it is one safe,effective bone graft substitute material for the filling of bone defects.BME-10X HAC forms bony connection after implanted in bone defects for 3-6 months without any adverse reaction.Chemical composition,crystallite size of the composite were similar to that of natural bone.Further research deserves to be executed.It is expected that the BME-10X HAC biomimetic composites will be an ideal bony repair substitutes.
