Objective: To study the influence of HBx gene to the apoptosis of the cell lines (L02, HepG2) and the relationship with the expression of XIAP.Methods: 1. Revivaled and cultivated HepG2 and L02 cell lines;2. pcDNA3.1-HBx was constructed and then be transfected cell lines (L02, HepG2) transiently;3. The expression of HBx in the cell lines was detected by RT-PCR;4. The expression of XIAP was investigated in mRNA levels using realtime RT-PCR;5. XIAP expression was detected by immunocytochemistry;6. The apoptosis of the cell lines was detected by flow cytometry.Results: 1. HBx gene can be detected in L02/pcDNA3.1-HBx and HepG2/pcDNA3.1-HBx;2. The morphous of L02/pcDNA3.1-HBx changed to fusiform from round, like HepG2;3. The mRNA expression of XIAP in L02 was about 3 times that in L02/pcDNA3.1-HBx cells; and in HepG2/pcDNA3.1-HBx was about 4 times that in HepG2 cells (P<0.01);4. The expression of XIAP protein in L02/pcDNA 3.1-HBx was lower than it in L02/pcDNA3.1 cells (P<0.05); and the expression of XIAP protein in HepG2/pcDNA 3.1-HBx was higher than it in HepG2/pcDNA 3.1 cells (P<0.01);5. The apoptosis rate of L02/pcDNA3.1-HBx was higher than that of L02/pcDNA3.1 cells (P<0.01); and in HepG2/pcDNA3.1-HBx cells, it was lower than in HepG2/pcDNA3.1 cells (P<0.05).Conclusions: HBx gene can enhance the apoptosis of normal hepatocellular and inhibit the apoptosis of hepatoma carcinoma cell. That role may be carried out by regulating the expression of XIAP.
