FeiJiNing Decotion Induced Apoptosis And Down Regulation Expression Of VEGF In Human Lung Adenocarcinoma Cell Line A549: An Experimental Study

Object: To explore the effect of FeiJiNing Decotion (FJN)extracts on proliferation,apoptosis and expression of Vascμlar endothelial growth factor (VEGF) in human lung adenocarcinoma cell line A549.Method: Culture A549 cell in vitro, the proliferate inhibit rate was evaluated by MTT assay, Apoptotic rate was determined PI/AnnexinV-FITC double staining , the morphological change of apoptotic cell was observed by fluorescence microscope and the growth state of cell was observed by inverted microscope.the expression of VEGF was detected by ELISA.Results: MTT assay showed that various dose teams can inhibit human lung adenocarcinoma A549 proliferation in a dose dependent manner.The IC50 is 200mg/ml,we selected four different Concentra -tion (30mg/ml,100mg/ml, 200mg/ml, 1100mg/ml ) as experimental concentration,and experimental time is 72h. Morphological study showed that ,with the dose of increasing ,the cell adhesion ability decreased,many cells shed from the culture bottle wall and floating in culture medium. The configuration changes the circle, the volume reduces, the cell spacing increases, and light refraction is stronger. Annexin V/PI double staining,The cell apoptosis was evaluated by flow cytometry.it shows that FJN decotion can deduce cell apoptosis,which increase with raised concentration .After the chi-square test,the cell apoptosis of four dose team is higher than control group, the difference is remarkable (P<0.05); the high dose team (1100mg/ml) is higher than control group (4.3%) obviously, p<0.01 .Annexin V/PI double staining,a typical set of changes such as cell shrinkage,cleavage of nuclear ,chromat in condensation , membrane blending and apoptotic body formation will be observed in fluorescence microsxope.All dose team can down regulation VEGF expression, and the degrees of their declines were in a dose - dependent fashion.when the Concentration is 200mg/mland1100mg/ml, th difference is significant compared with control group.With the increasing of concentration , inhibit rate of proliferation become larger ; We may induce the A549 cell to Apoptosis, when the extract concentration achieved 1100mg/ml, apoptotic rate is higher than the control group obviously (44.8%: 3.4%, P<0.01);Various dose teams may down regμlation VEGF expression, but the middle,high dose team may cause it decline significantly (P<0.01) , The degrees of their declines were in a dose - dependent fashion.Conclusion:FJN has anticarcinogenic function ,and the possible mechanic of which are induced apoptosis and down regμlation expression of VEGF.
