Objective: 1,To study the effect of thalidomide combined with cisplatin(DDP) on the proliferation and apoptosis of human lung carcinoma cell line A549, and to judge if there are synergistic or additive pattern between the two drugs; 2,To study the effect of thalidomide on the express of VEGF and BFGF in lung cancer cellMethods: A549 cells were added in RPMI-1640 supplemented with 10% FBS, 100u/ml penicillin, and 100u/ml streptomycin, cultured in a humidified 5% CO2/95% air atmosphere at 37°C. 1,While cells proliferated in logarithm, the effect of thalidomide combined with cisplatin on the proliferation of A549 cells was evaluated by MTT reduction assay, and the Q value was calculated to judge the combined effect. 2,A549 lung cancer cell cultured with DDP(2mg/l,4mg/l) with or without thalidomide for 48h, the apoptosis was detected by Propidium Iodine(PI) flow cytometry. 3,To observe the effect of thalidomide on angiogenesis enhancing factor such as vessel endothelium growth factor (VEGF ) and bsic fibroblast (BFGF) synthesized by A549 cells with immuocytochemistry.Results: 1,Both thalidomide and cisplatin could inhib it A549 cells proliferation in a concentration-dependent pattern(r=0.987 and 0.547, respectively; p<0.01, respectively). When cisplatin alone,2mg/L could significantly inhibit cells proliferation. When thaalidomide alone,7.5mg/L could significantly inhibit cells proliferation. While thalidomide(7.5mg/L) combined with cisplatin(0.5 mg/L), A549 cells proliferation was inhibited,While thalidomide combined with cisplatin, A549 cells proliferation was significantly inhibited( P<0.05)),compared with cisplatin. In a certain concentration range, the combination of the two drugs can increase inhibition rate in a time-dependent pattern( r=0.808 P<0.01). 2. Flow cytometry show that apoptosis maybe observed 48h after administration of thalidomide combined with cisplatin in A549 cells. Apoptosis proportion will be higher with the increase concentration of thalidomide combined with cisplatin. 3.Thalidomide could inhibit VEGF and BFGF albumen expression in A549 cells ( t=459.34,p <0.05 ;t=543.83p<0.05). Conclusion: 1. Thaldomide combined with cisplatin could enhance the inhibitory effect on the proliferation of human lung adenocarcinoma cell line A549, when the concentration of cisplatin is equal to or more than 0.5mg/L, Additive effect between the two drugs was found; 2. Thaldomide combined with cisplatin could increase the apoptosis rate,It indicates that anticancer mechanism may be closely associated with apoptosis. 3. Thaldomide can obviously inhibit A549 lung cancer cells such as VEGF and BFGF.
