Experimental Study Of Yi Qi Bu Fei Formula On Lung Function Decrease In Rats

Objective:On the basis of inflammation theory,to explore whether "Yi Qi Bu Fei Formula (YBF)" could accommodate the levels ofTNF-α,ET-1,MIF to improve acute lung injury through the rats experiment,to provide a new clinical treatment.Methods:The rats were randomly divided into five groups:YBF,dexamethasone(DXM), YBF+DXM,positive control,negative control.YBF and YBF+DXM were ntragastric administrated by formula(2ml/d) for 3days,while the other groups used normal sodium;DXM and YBF+DXM were intravenous injected by DXM(1mg/kg) while the other groups used normal sodium.Every groups were intravenous injected by oleic acid 1h after corresponding manipulation on 3rd day.The action of rats,lung ratio, TNF-α,ET-1 and MIF levels in lung tissue,lung histologic examinations were conducted after model establishing.Results:Compared to negative control group,lung ratio,TNF-α,ET-1 and MIF levels in other groups were raised in varying degrees,the differences among them were significant (P＜0.05).The lung ratio was highest in positive group,while lowest in negative group,the differences among drug-using groups were not significant(P＞0.05). TNF-α,ET-1 and MIF levels in all groups were consistent with normal distribution and homoscedasticity,and levels were reduced compared to positive group,while difference of ET-1 level among drug-using groups was significant Histologic changes of alveoli fibrin effusion was significantly improved in YBF group,while interval widen change was improved in both YBF and YBF+DXM groups,the interval difference was not significant.Conclusion:1.The ALI model was established successfully in rats by injection of oleic acid. YBF and DXM both have prophylactic and therapeutical effects on ALI in rats, and there is no synergistic effect between them.2.YBF protects the acute rats lung injury induced by oleic acid by inhibiting TNF-α, ET-1 and MIF.
