| Angelica is the dried root of Angelica sinensis (oliv) Diels,and is commonly clinical drug. Traditional Chinese Medical (Veterinary) considers that angelica is one of the best drug for treatment of the blood diseases. It is sweet and pungent in taste, and warm in nature, acts on the liver, heart and spleen meridians and has the effects of invigorating the blood, promoting blood circulation, regulating menstruation, alleviating pain, moistening the bowels, and relieving constipation. Modern pharmacology further studies proved that angelica had the effects of promoting blood circulation by removing blood stasis, invigorating the blood and alleviating pain. It was also found that angelica had the effects of anti-inflammation, scavenging the free radical (O2-), protecting liver, cholagogue, protecting kidney and enhancing immune function. This study prepared water and alcoholic extracts from angelica with synergistic extraction by ultrasonic-microwave. In order to study the effect of water and alcoholic extracts from angelica on cleaning free radical , anti-lipidperoxidation in vivo and improving anti-oxidation in vitro. This study is aimed at providing scientific references for processing, pharmacological effects and application of angelica.1.The effect of different extracts from angelica were determined on cleaning·OH free radical by producing hydroxyl with Fenton reaction. Experimental results suggested that water and alcoholic extracts from angelica had significantly the effect of scavenging·OH free radical in 6~50mg/ml and 12.5~200mg/ml respectively. Clearance rate showed a concentration-dependant manner, the scavenging effect of water extract was superior to alcoholic extract from angelica for the same concentration.2.The effect of different extracts from angelica were determined on cleaning O2- free radical with the autoxidation of pyrogallol model. respectively. Experimental results suggested that water and alcoholic extracts from angelica had significantly the effect of scavenging O2- free radical in 12.5~200mg/ml, the scavenging effect of water extract was superior to alcoholic extract from angelica for the same concentration.3.The effect of different extracts from angelica were determined on MDA spontaneous formation in liver homogenates of mice by TBA colorimetry. Experimental results suggested that water and alcoholic extracts from angelica can significantly inhibit MDA spontaneous formation in homogenates of mice liver in 25~200 mg/mL, Inhibitory effect increases gradually with increasing of drug concertration, inhibitory rate showed a concentration-dependant manner,the inhibitory effect of alcoholic extract from angelica was better than water extract from angelica for the same concentration.4.The effect of different extracts from angelica were determined on erythrocytic membranous lipid peroxidation and hemolysis induced by H2O2 were determined with spectrophotometers. Experimental results suggested that extracts from angelica can significantly inhibit erythrocytic membranous lipid peroxidation and hemolysis in 25~200 mg/mL. Inhibitory effect increases gradually with increasing of drug concentration, inhibitory effect of different extracts from angelica showed a concentration-dependant manner, the inhibitory effect of alcoholic extract from angelica for erythrocytic membranous lipid peroxidation induced by H2O2 was superior to water extract from angelica for the same concentration, the inhibitory effect of water extract from angelica for erythrocytic hemolysis induced by H2O2 was better than alcoholic extract from angelica for the same concertration.5.The effect of water extract from angelica were determined on MDA,SOD and GSH-PX by TBA colorimetry, nitrite and DTNB methods. Experimental results suggested that water extract of angelica can inhibit MDA spontaneous formation in homogenates of mice liver and kidney, bring down contents of MDA, significantly increase activity of SOD and GSH-PX. The effect was gradually increasing for reducing contents of MDA, improving activity of SOD and GSH-PX with increasing of drug concentration. Water extract of angelica had effects of oxidation resistance and improving activity of antioxidase. |
