On the basis of the experimental study of the EP tumor cells originating from lymphocytes, the reverting phase variation process and the mechanism thereof had been further explored. The routine cellular staining technique, AFM scanning technique, continue culture and indirect immuno-fluorescent method, as well as the cellular transformation function test and BrdU incorporation test and animal inoculation test had been employed to explore the morphological changes, topological features, cultural characteristics and CD3 and CD40 expression, as well as the cellular transformation function and cellular proliferation, also, the ability of growing into tumors respectively. The results showed that during the evolution at the inoculation local the tumor with the diameter less than 1 cm, or more than 2 cm exhibited smaller ratio of cells positive in CD antigens to those of negative. Whereas, those with the diameter of 1 cm to 2cm, the ratios were higher. On the other hands, with the times of generations increase, the proliferation of EP tumor cells slowed down, and ultimately stopped to be the cells that did not multiply anymore, and the characteristics of morphology, topological under atomic force microscope, and the surface CD3 and CD40 antigen expression, as well as the cellular transformation and proliferation function under the stimulation of LPS, Con A, PHA, PWM and also the ability of growing into tumors, all are tending to be similar with that of lymphocytes, which disclose a possibility that the tumor cells possess a potential tendency of reverting to their original cells under certain environmental conditions. This may be the vary mechanism underlying the clinical self-regression of tumors and thereafter this experimental research supply a experimental mimic method for the research of tumor regression and the cancer cell reversion, and a experimental basis for theoretically elucidating the mechanism of eliminating the tumor in the regulating treatment with biological wave regulating factors. Therefore this study may be a help to prompt the birth of the new method of anticancer, which take the phase variation of tumor cell tending to revert to original cells as the theoretical foundation.
