Experimental Study On Effect Of HPT,HPG Axle With Mouse Depression Model And The Optimum Scheme Of Prescription For Xindaxin Formula

Objective:To screen and evaluate antidepressant activities of clinically empirical Xindaxin Formula designed by famious tranditional Chinese medicial doctor Zhou Zhongying,including the determination of ideal herb combining and dosage level,and influence research of mouse HPG and HPT axle.Methods:(1)To utilize error frequency of Learned Helplessness km mousse depression model avoiding darkness ethology to be the pharmacodynamics index,and calculate the best dosage level in each herbs of Xindaxin Formula by pharmacology software DAS;(2)To research Xindaxin Formula's effection of the HPT axle by observing ethology index and blood-serum' T₃,T₄ of Learned Helplessness model,To study the Xindaxin Formula's therapy influence on HPG axle and genital system by detect ethology index and blood-serum's E₃,LH,PRL of stress female km mouse model.Results:(1)Among the herbs of original formula(XDXF),herb B showed the strongest effect to decrease elusion times,while herb A,C can increase the anti-depression effect of herb B;(2)Compared with the normal group,model group mouses'weight obviously descented,activities frequency descended,elusion times increased.After treated by Xindaxin Formula,the model group got heavier,physical condition got better,and activities frequency increased,increased PRL level descended obviously(P＜0.05),decreasedT₃,T₄ increased obviously,vagina caduceus cell cornification exponent,LH,E₃ showed increasing tendency,manifested great difference compared with the Fluoxetine group,lessen depression of the model group,showed no significant difference compared with the Fluoxetine group(P＞0.05).Conclusions:(1)Xindaxin Formula is composed of based on an ideal model and the optimum scheme of prescription can give the best therapeutic effect on learned helplessness mouse;(2)Xindaxin Formula can obviously release depression condition of the mouses,the mechanism is concerned with the accommodating the mouses humen HPG and HPT axle,it provides the clinical depression therapy rationale of Xindaxin Formula.
