Extramammary Paget Disease(EMPD) is a common cutaneous malignant carcinoma relatively,it isn't very clear for EMPD's characteristics of growth and invasion.The purpose of the subject is to study the role of E-cadherin,Matrix Metalloproteinase 2,Membrane-type 1 Matrix Metalloproteinase and Hypoxia-inducible Factor-1 alpha in the process of invasion in primary EMPD.First,we detected the expression of E-cadherin(E-cad,an important adhesion molecule in the process of tumor invasion) in EMPD by immunohistochemical methods.The expression of E-cad in primary EMPD compared with that in normal skin was downregulated. There were significant statistical difference about the expression of E-cad between the group of normal skin and the groups of primary EMPD in situ/invasion;however,which didn't show significant statistical difference between the group of primary EMPD in situ and the invasive group.Second,we detected the expression of Matrix Metalloproteinase 2(MMP-2), Membrane-type 1 Matrix Metalloproteinase(MT1-MMP) and Hypoxia-inducible Factor-1 alpha(HIF-1α) in EMPD by immunohistochemical methods.The expression of MMP-2, MT1-MMP and HIF-1αin primary EMPD compared with that in normal skin were upregulated.MMP-2 and MT1-MMP had statistical significant positive correlation,similar results were found in MMP-2 and HIF-1αexpression.MT1-MMP and HIF-1αexpression demonstrate the same results.There were significant statistical difference about the expression of MMP-2,MT1-MMP and HIF-1αbetween the group of normal skin and the groups of primary EMPD in situ/invasion,while there were no significant statistical difference about the expression of MMP-2,MT1-MMP and HIF-1αbetween the group of primary EMPD in situ and the invasive group.Those results suggest that downregulation of E-cad and upregulation of MMP-2, MT1-MMP and HIF-1αshould be correlated to the development of EMPD,and which create conditions for the invasive growth of EMPD.Ultimately,many other factors may participate in the invasive growth of EMPD.
