Part 1 Establishment of rat cell line C6 brain glioma models with stereotaxisObjective To establish brain glioma models of C6 cells in rats with different volumes to compare the growth patterns of the intracranial tumors. Methods To prepare cell suspension of 1.0×106 C6 cells/10μl to stereotaxically implant into the right caudate nucleus with different volumes ( 5μl,10μl,20μl,40μl,80μl,160μl). To observe the living states of rats. To investigate the volumes,metastasis,histopathology,expression of C-erbB1 and S-100B proteins of tumors. Results The formation rates of brain tumors were 60%(5μl),93%(10μl),100%(20μl),93%(40μl),87%(80μl),33%(160μl). The tumors were highly cacoethic, with remarkable nuclear polymorphism. Immunohistochemical staining showed C-erbB1 and S-100B proteins were strongly positive in rat tumors. Conclusions The rat C6 glioma models can be successfully established with implantation volumes from 10μl to 80μl. The tumors of rats resembles human glioma in histopathological and morphology features.Part 2 the Relationship between Cerebral Edema and erb-B1 Protein Expression in Glioma Rats Treated by Co60Objective To observe the relationship between cerebral edema and erb-B1 protein expression in glioma rats treated by Co60. Methods Built glioma rats model. After 15 days, treated the control group with Co60, treated the experiment group with Co60 and ZD1839. Immunohistochemical technique SP stain checked erb-B1 protein expression. Computer image analysis. Results The level of erb-B1 protein expression were significantly different between the serious edema rats and the none edema rats in control group or experiment group(P<0.05), the blank group and the edema rats of control group(P<0.05), the blank group and the edema rats of experiment group(P<0.05). The serious edema percents were significantly different between the control group and the experiment group (P<0.05). Conclusions The mechanism of radioactive cerebral edema is tightly concern with erb-B1 protein expression after Co60 therapy.Part 3 The relationship between the differences of the therapy of radioactive cerebral edema with methylprednisolone and Dexamethasone and the transcription level of erb-B1Objective To study the effect of methylprednisolone(MP)and Dexamethasone (DXM) to radioactive cerebral edema and the transcription level of erb-B1. Methods One hundred male Wistar rats were divided into 5 groups atrandom (n=20). Group A was irradiated with 30 mg/kgMP, group B was irradiated with 10 mg/kg MP, group C was irradiated with 5 mg/kg DXM, group D was irradiated without glucocorticoid(GC), group E was normal control. Planted glioma cells into brains of rats of group A,B,C,D and E, and treated groups A,B,C,D with Co60 after 15 days. All groups give GC or normal saline beginning 3d before and ending 6d after irradiation, The water content of the brains and the transcription level of erb-B1 were measured 6d after irradiation. Results The water content of the rats brain :There was significant difference between group D,E and group A,B,C(P<0.05) ,group D and group E(P<0.01), group C and group A,B(P<0.05);There were no significant difference between group A and B(P>0.05). The level of erb-B1 mRNA: There were significant difference between group D,E and group A,B,C (P<0.05), group D and group E(P<0.01), group C and A,B(P<0.05). There were no significant difference between group A and B(P>0.05). Conclusions MP and DXM can both protect the radiation-induced injury and brain edema changes by inhibiting the transcription level of erb-B1 mRNA in rats, and the MP is more efficient than DXM.
