Objective: Avascular necrosis of femoral head is a a kind of disease which manifested as femoral head necrosis because of damages to the blood supply of femoral head. The bone morphogenetic proteins (BMPs) are a family of growth factors that regulate the development of bone. Danshen is one of the traditional Chinese herbs,which has effects of improving microcirculation.Our objective is to investigate the effects of salvia injection in accelerating the progress of repairing of femoral head necrosis in rabbits and in accelerating the progress of revascularization.Methods: 24 Models of Femoral head necrosis were established which were divided into 3 groups: Group A: BMP-autogenous bone and salvia injection group; Group B: BMP-autogenous bone group and Group C: natural repair group. We studied the osteogenic function by alkaline phosphatase, calcirm, phosphorus and histomorphology changes during the progress of repairing of femoral head necrosis. We investigate the revascularization in the progress of repairing of femoral head necrosis by immunohistochemistry. The microvessel density (MVD) in the specimens was detected by CD34 staining.Results: After 12 weeks' therapy, the function of osteogenic in group A was superior to group B and group C significantly(P<0.05). These results in group B were better than those in group C(P<0.05). The blood supply of femoral head in group A was obviously better than that of group B and group C(P<0.05). While there were no significant difference between groupB and group C(P>0.05). The pathological section shows the bone trabecular was distributed in all over the filling areas in group A. Disordered new born bone trabecula was found in group B. In group C, the bone trabecula was sparse and thin. Immunohistochemical analysis found that the number of capillaries in group A was much more than that in group B and group C(P<0.05). While there was no significant difference in the number of capillaries between group B and group C(P>0.05).Conclusion: Salvia injection can increase the number of capillaries in femoral head, accelerate the process of revascularization and improve the blood supply of femoral head. Salvia injection combined with bone morphogenetic protein could increase calcium content, phosphorus content, alkaline phosphatase activity and enhance the effect of BMP on repair of bone defect.
