Objective and significanceDerinatosis of melanin dysbolism(chloasma,vitiligo,etc) which has a high incidence of disease makes a bad influence on patient's life.But people have not get its exact influence factor and its pathogenesis by now.As is well-known to us,zoopery is the foundation of medicine's development,and experimental animal is one of the most important carrier of medicine study.There were many research had down in derinatosis of melanin dysbolism,but most report were about how to make its animal model and what the action of monophenyl oxidase,so there were no report of how to choose the appropriate animal and whether have other pathogenesis.Actually,different experimental animal has different physiologic characteristics, and choosing appropriate experimental animal plays a great role in making a succeed animal model,so we should acquired some theory evidence.In now, many research have cinfirm that derinatosis of melanin dysbolism caused by dysbolism of skin's melanin,and melanin originates in melanocyte.Under the function of monophenyl oxidase which are containd in melanocyte, beta-parahydroxy-phenylpropionic acid is oxidated and reseted,and finally becomed melanin.As is approved by recent reports, melanocyte of scarfskin can produceα-melanocyte-stimulating hormone(α-MSH) which can combine with melanin 1 receptor,after that,they can activate the expression of monophenyl oxidase,and accelerate melanocyte to secrete melanin.Soα-MSH plays a great role in the production of melanin.In order to clarify how to choose the appropriate animal and discuss the accurate pathogenesis,we decide to make research of observing the expression of melaninα-MSH in white- mouse of Kun ming,black-mouse of C57BL/6J ,white-guinea pig and brown-yellow- guinea pig's scarfskin.Methods and contentFemale Kun ming mouse ,C57BL/6J mouse,female FMN guinea pig and HARTLY guinea pig were selected as laboratory.According to their different genus,made the animals into four groups.Injected 3% pentobarbital of 45mg/kg into these animals after a week,and scraped their back hair,and cutted many pieces of skin as 2cm×2cm from animal's back,then made the samples into paraffin section.1 The expression of melanin in four different animal's scarfskinChose the same amount of sample from each group,and used the technique of immunohistochemical staining to observe the melanin.The result was analyzied according to the positive cell population,the sectional area of cell and gray value,and photoed positive cell under optical microscope.2 The expression ofα-MSH in four different animal's scarfskinChose the same amount of sample from each group,and used the technique of immunohistochemical staining to observe theα-MSH. On base of study on the expression of melanin andα-MSH in four different animal's scarfskin,we used SPSS13.0 to analyzied these experimental date.Result1 The expression of melanin in four different animal's scarfskinThere were almost no melanin positive expression in group of both female Kun ming mouse and female FMN guinea pig.The positive expression of melanin about the positive cell population,the sectional area of cell and gray value in group of HARTLY guinea pig are better than in the other groups' (p<0.01), and those in group of C57BL/6J mouse are better than in the other two white groups (p<0.01),and there are no different in the two white groups(p>0.05).2 The expression ofα-MSH in four different animal's scarfskinThere were almost noα-MSH positive expression in group of both female Kun ming mouse and female FMN guinea pig.Theα-MSH positive expression rate was 20% in group of C57BL/6J mouse while 55% in group of HARTLY guinea pig.The comparison between C57BL/6J mouse and HARTLY guinea pig has significant difference(p<0.01).Conclusions1 Female Kun ming white mouse and female FMN white guinea pig are not suit for experiment of observing melanin and a-MSH;they can be used for experiment of chloasma or other pigment-added disease.2 C57BL/6J black mouse is suit for experiment of observing folliculi pili's mslanin orα-MSH.3 HARTLY brown-yellow guinea pig is suit for experiment of observing scarfskin's mslanin orα-MSH.4 HARTLY brown-yellow guinea pig's ability of producing melanin is better than C57BL/6J black mouse's,so HARTLY brown-yellow guinea pig is more suit than C57BL/6J black mouse in making experiment of observing melanin orα-MSH. 5α-MSH can be a target of studying derinatosis of melanin dysbolism.
