Objectives:To explore the relationship between pathogenesis of premenstrual syndrome (PMS) and progesterone,glutamic acid (Glu) andγ–aminobutyric acid (GABA),seek for the influence of the drug and probe into its mechanism by observing the changes of progesterone and amino acid.Methods: Make sure the estrum of rats by observing the vagina cell shape. The PMS model rats with liver-qi invasion were induced by multiple emotional stimulations,Baixiangdan capsule was adminisiterd before and after PMS induction. The content of progesterone in serum and different brain regions which include the hypothalamus,the marginal,the cotex and hippocampus of rats was detected by radio-immune assy;and a fluoroscopy high performance liquid chromatographic method (HPLC) was developed for the detection of GABA and Glu.Results:Compared with the control group,the progesterone,the content of Glu and GABA in the serum of liver-qi invasion model rats reduced markedly(P﹤0.05,P﹤0.01,P﹤0.01),and they all increased markedly after medication. In different regions,compared with the control group,the content of progesterone of the modle rats reduced markedly(P﹤0.01),the content of progesterone in regions such as the hypothalamus,cotex and the hippocampus increased markedly after medication. The content of progesterone in the marginal increased but insignificantly. In the hypothalamus,the content of Glu and GABA of the model rats reduced markedly(P﹤0.01,P﹤0.05),and they increased after medication but Glu insignificantly. In the marginal,the content of the two amino acid reduced markedly(P﹤0.01),and they both increased after medication(P﹤0.05). In the cotex,the content of the two amino acid increased(P﹤0.01,P=0.0551),GABA decreased after medication(P﹤0.05),Glu decreased but insignificantly. In the hippocampus,the two amino acid increased(P﹤0.01),and GABA increased significantly after medication(P﹤0.01)while Glu changed insignificant.Conclusions:PMS with liver-qi invasion was in close relation to the changes of the content of progesterone,Glu and GABA in serum and different brain regions. Baixiangdan capsule can regulate the content of P,Glu and GABA in the organize of the PMS model rats with liver-qi invasion in different degrees,that might be one of the most important action mechanism to cure PMS of the medicine. The changes of the content of progesterone,Glu and GABA in serum and different brain regions might be an important cause resulting in PMS with liver-qi invasion.
