OBJEVTIVE:To study influence of exercise training on the structure and function of skeletal muscles of rats with chronic excessive drinking.METHODS:Totally 30 male Wistar rats(200—220g) were selected and randomly divided into 3 groups,control group,alcohol group and exercise group,ten animals were involved in each group.The ethanol group and exercise group was administered ethanol(50%)10g·kg-1·day-1 in 10 weeks.The control group was lavaged isovolume normal saline.Meanwhile,from 7th week the rats of the exercise group was begun to be administered with swimming training.Measurements of weight of each rat were performed every week.After 10 weeks the experiment made use of RM-6240 system, observe construction function of gastrocnemius and soleus muscle by suscitation of ischiadic nerve in vivo.The experiment observed morphous and distribution of skeletal muscles fiber,and measured cross section area of the fibers.A quantitative analysis was carried out for total protein skeletal muscles.RESULTS:1.The construction function of skeletal muscle of mice:(1) the latency of single twitch of gastrocnemius of the alcohol group was prolonged compared with control group,and contraction extent was significantly decreased,1/2 relaxation time of single twitch also prolonged(P<0.05);the latent period of tetanus was prolonged, contraction extent of tetanus was decreased,and fatigue index number was increased significantly.(2) Indexes of construction function of soleus of three groups did not appear disparity conspicuously(P>0.05).2.The cross section area of the fibers of skeletal muscle of mice:(1) The cross section area of the fibersⅠof gastrocnemius of the alcohol group did not appear disparity conspicuously with control group(P>0.05), The cross section area of the fibersⅡa andⅡb of gastrocnemius of the alcohol group was diminished compared with control group(P<0.05).The cross section area of the all types fibers of gastrocnemius of exercise group did not appear disparity significantly with alcohol group(P>0.05).(2) The cross section area of all types fibers of soleus of three groups did not appear disparity conspicuously(P>0.05).3. Quantitative of total protein of skeletal muscles:(1) Quantitative of total protein of gastrocnemius reduced significantly with control group(P<0.05).Quantitative of total protein of gastrocnemius of exercise group did not appear disparity significantly with control group(P>0.05).(2) Quantitative of total protein of soleus muscle of the three groups did not appear disparity conspicuously(P>0.05).CONCLUTION:Chronic excessive drinking can damage the structure and function of gastrocnemius of rats,and regular exercise training can lighten it.
