Objective:The paper observes the effects on expression of estrogen receptor alpha (ERα),nerve growth factor(NGF),Interleukin-2(IL-2),androgen receptor(AR) protein and mRNA in the ovariectomized rat's hypothalamus-pituitary-spleen by supplying soybean isoflavone(SI).Discuss the effects and the mechanism of interaction of these four material in the ovariectomized rat's hypothalamus,pituitary and spleen by supplying SI.Research the effect of SI on the flora in intestinal tract by separating the content of intestinal tract.Method:50 young female rats were divided into sham operated group and ovariectomized group at radom.In the ovariectomized group,rats were supplied with SI respectively in high,middle and low control(solvent) dosage.In the second week and the sixth week,5 rats per group were killed for experiment.Researched the expression and distribution of ERα,NGF,IL-2,AR protein and mRNA in the hypothalamus, pituitary and spleen after SI treatment in different dosage by using technique of immunohistochemical SABC and oligonucleotide probe hybridization in situ.Count plate method was used for the study of the flora in intestinal tract.Results:①The protein and mRNA of ERα,NGF,IL-2,AR distrubuted in arcuate nuclei,dorsomedial nucleus of hypothalamus,necleus hypothalamicus ventromedialis,periventricular nucleus and periventricular hypothalamic nucleus,which showed that ERα,NGF,IL-2, AR played a wide role in the hypothalamus.The four productions mainly located in nucleus and neurite,a little in plasma and cytoplasmic membrane,which showed that the genetic transcription by endogenous estrogen was controled mainly by classic nuclear receptor way in hypothalamus and partly by nongenomic mechanism.The general tendency of the intensity and quantity of ERα,NGF,IL-2,AR in hypothalamus of OVX rat is significantly decreased,while ascended in different degree after SI treatment.In the 6H group,the intensity and quantity of these productions can basically reach the level of those in the sham-operation group;There's little augment in the 6L group;Those in the 6M group were found between the two;Only the 2H group in the 2 week groups were found a little augment.It showed that SI could upgrade the level of estrogen in vivo and promote the expression of ERα,NGF,IL-2,AR.It showed that SI could upgrade the level of estrogen in vivo,promote the expression of ERα,NGF,IL-2,AR.and play an important role in sustaining and protecting the structure and function of hypothalamic neurons,depending on the dosage and time.Furthermore,the similarity of these four material's changing tendency implied that they were correlated and cooperated during the course in effect of SI on hypothalamus.②The protein and mRNA of ERα,NGF,IL-2, AR distrubuted in antepituitary,hypophysenmittellappen,hypophysenhineerilappen.The four productions mainly located in nucleus,seldom in plasma and cytoplasmic membrane,which showed that ERα,NGF,IL-2,AR joined in the progress of effecting the physiologic function of pituitary.The general tendency of the intensity of ERα,NGF, IL-2,AR and the number of positive cells in pituitary of OVX rat is significantly decreased,especially 6C group comparing with 2C group.While ascended in different degree after SI treatment.In the 6H group,the intensity and quantity of these productions can basically reach the level of those in the sham-operation group;There's little augment in the 2 week groups and 6M,6L group,and the changing tendency of the distribution and intensity was nearly the same.It showed that SI played an important role in sustaining and protecting the structure and function of pituitary,depending on the dosage and time.Furthermore,SI was correlated and cooperated during the course in effect of SI on pituitary.③ERα,NGF,IL-2,AR were distributed widely in spleen,espacially in red pulp,little in white pulp.The distribution area of mRNA was the same with those four proteins,only different in quantity,which meant that the expression of mRNA was more than proteins,and implied that ERα,NGF,IL-2,AR joined in the progress of effecting the immuno-regulation of spleen.The general tendency of the intensity of ERα,NGF, IL-2,AR and the number of positive cells in spleen of OVX rat is significantly decreased. This degression was more significantly in 6Cgroup.In the 6H group,the intensity and quantity of these productions can basically reach the level of those in the sham-operation group;There's little augment in the 2 week groups and 6M,6L group.It showed that SI could promote the expression of NGF,IL-2,AR via ERα,depending on the dosage and time.The changing tendency of the distribution and intensity was nearly the same.It implied that SI was correlated and cooperated during the course in effect of SI on spleen.④The distribution areas of ERα,NGF,IL-2,AR protein and mRNA in hypothalamus, pituitary,spleen nearly resembled.It suggested that there may be coexpression area in hypothalamus,pituitary,spleen.These materials could adjust their coexpression sites in the three organs by interaction.The changing tendency of the distribution and intensity was at equal pace,which showed the effect of SI on hypothalamus-pituitary-spleen and ERα,NGF,IL-2,AR were co-regulation and cooperated.⑤SI could suppress the growth of Escherichia coli selectively and promote the growth of Streptobacterium plantarum, which suggested that SI could adjust the microecological balance of flora.
